老年肝郁失眠大鼠模型的建立及舒郁安神方作用的研究

【摘要】: 目的:建立老年肝郁失眠大鼠模型,探討舒郁安神方治療老年肝郁失眠的作用機(jī)制 方法:采用腹腔注射D-半乳糖致亞急性衰老,進(jìn)行形態(tài)、體重觀察,并測定腦組織SOD、MDA含量;夾尾刺激致肝郁,并進(jìn)行形態(tài)觀察以及血漿6-keto-PGF_(1a)、TXB_2的檢測;改良多平臺水環(huán)境法致失眠,并進(jìn)行形態(tài)觀察和錄像監(jiān)控,從而建立老年肝郁失眠大鼠模型。通過延長戊巴比妥鈉睡眠時間實驗和戊巴比妥鈉閾下催眠劑量實驗,以舒樂安定為對比,研究舒郁安神方的促眠作用。Morris水迷宮中的定位航行和空間探索實驗用于測定舒郁安神方對老年肝郁失眠模型大鼠空間學(xué)習(xí)記憶能力的影響。舒郁安神方對老年肝郁失眠模型大鼠腦組織Glu及GABA含量的影響,則采用高效液相法分別測定額葉皮質(zhì)及下丘腦部位Glu、GABA的含量,并與空白對照組、證候模型組及舒樂安定組作比較。 結(jié)果:衰老造模后,老年組、模型組大鼠體重增長緩慢,六周后體重與空白組對比,差異有顯著性(P<0.01);腦組織SOD、MDA含量與空白組對比,差異有顯著性(P<0.01),而老年組與模型組之間差別無統(tǒng)計學(xué)意義。肝郁造模后,肝郁組、模型組血漿TXB_2、6-keto-PGF_(1a)含量以及6-keto-PGF_(1a)/TXB_2比值與空白組對比,差異有顯著性(P<0.01),而肝郁組與模型組之間差別無統(tǒng)計學(xué)意義。睡眠剝奪實驗可觀察到模型組與失眠組大鼠的總睡眠量明顯減少,而空白組無此變化。促眠實驗結(jié)果可見,與空白對照組比較,舒郁安神方組可協(xié)同戊巴比妥納延長小鼠睡眠時間(P<0.05)、提高小鼠戊巴比妥鈉閾下劑量時的入睡率(P<0.01);陽性對照藥安定的作用較舒郁安神方的作用更加顯著,均有顯著差異(P<0.01)。大鼠定位航行實驗中,模型組與安定組第3天、第4天的逃避潛伏期均較對照組和舒郁組所用的時間長,其兩天平均成績也明顯低于對照組和舒郁組,差異均具有顯著性(P<0.01),安定組與舒郁組之間則有顯著性差異(P<0.01)。大鼠在空間探索實驗中,模型組、安定組與對照組、舒郁組相比,20%邊緣區(qū)域所占比例明顯增多,40%邊緣區(qū)域及中心區(qū)域所占比例明顯減少,差異均具有顯著性(P<0.01),而模型組與安定組之間的差異及對照組和舒郁組之間的差異則無顯著性(P>0.05)。模型組大鼠額葉皮質(zhì)及下丘腦部位Glu的含量與空白對照組相比,均顯著升高(P<0.01);安定組大鼠各腦區(qū)Glu含量與模型組相比,均明顯下降(P<0.01),舒郁組與空白對照組相比,各腦區(qū)Glu含量顯著升高(P<0.01);與安定組對比,舒郁組在額葉皮質(zhì)部位的含量呈現(xiàn)升高,差異有統(tǒng)計學(xué)意義(P<0.05);而在下丘腦部位的含量則明顯升高(P<0.01)。與空白對照組相比,模型組、安定組大鼠額葉皮質(zhì)及下丘腦部位GABA的含量顯著升高(P<0.01);舒郁組與空白對照組相比,含量有所升高,差異有統(tǒng)計學(xué)意義(P<0.05),與模型組對比,舒郁組在腦區(qū)各部位的含量均呈現(xiàn)顯著下降(P<0.01);舒郁組與安定組的各腦區(qū)含量比較,額葉皮質(zhì)部位含量表現(xiàn)為下降(P<0.05),下丘腦部位則下降明顯(P<0.01)。 結(jié)論:老年肝郁失眠大鼠模型的建立是較為成功的。舒郁安神方具有較好的促眠作用,對老年肝郁失眠大鼠模型的學(xué)習(xí)記憶能力具有一定的保護(hù)作用,而調(diào)節(jié)GABA、Glu兩種神經(jīng)遞質(zhì)的含量比值則可能是其治療老年肝郁失眠的神經(jīng)生物學(xué)作用機(jī)制之一。
[Abstract]:Objective: to establish a rat model of senile liver depression and insomnia, and to explore the mechanism of Shu Yu Anshen prescription in the treatment of senile liver depression and insomnia.
Methods: intraperitoneal injection of D- galactose induced aging, morphological observation, weight, and determination of brain tissue SOD, MDA content; tail pinch induced liver, and to observe the morphology and plasma 6-keto-PGF_ (1a), TXB_2 detection; modified multiple platform method caused by insomnia, and to observe the morphology and video monitoring, so as to establish a rat model of insomnia in elderly depression. By prolonging the sleeping time induced by pentobarbital sodium experiment and pentobarbital sodium subthreshold hypnotic dose experiment with estazolam for comparison, research and promotion of Shuyu positioning navigation and space effect of.Morris water maze sleep experiment for determination of Shuyu Anshen recipe learning the memory ability of aged rats. Liver depression and insomnia effect of Shuyu Anshen Decoction on Glu and GABA in the brain tissue of Elderly Rats Liver Depression Insomnia model, using HPLC and cortex were measured. The hypothalamus Glu, GABA content, and with the control group, syndrome model group and the diazepam group for comparison.
Results: the aging model, elderly group, body weight of rats in model group increased slowly, weight compared with the control group after six weeks, there was significant difference (P < 0.01); brain tissue SOD, MDA content compared with the control group, there was significant difference (P < 0.01), but no significant difference in the elderly between the group and model group. After the model of liver qi stagnation syndrome group, model group, plasma TXB_2,6-keto-PGF_ content (1a) and 6-keto-PGF_ (1a) /TXB_2 ratio compared with the control group, there was significant difference (P < 0.01), but there is no significant difference between depression group and model group. Sleep deprivation can be observed the total amount of sleep and insomnia model group rats were significantly decreased, while the control group no significant changes. Hypnogenetic experimental results show that compared with the control group, and group of Shu Yu can cooperate with sodium pentobarbital sleeping time in mice (P < 0.05), improve the pentobarbital sodium in small doses sleep 鐜,
