小鼠多潛能成體干細(xì)胞生物學(xué)特性及向樹突狀細(xì)胞分化的研究

【摘要】: 越來越多的研究發(fā)現(xiàn)成體干細(xì)胞具有跨系甚至跨胚層分化的特性,即成體干細(xì)胞具有可塑性,而對(duì)于可塑性的原因和機(jī)制,目前更多的學(xué)者認(rèn)為在成體組織中可能存在著原始的多潛能干細(xì)胞,我們命名為胚胎后亞全能干細(xì)胞。對(duì)于亞全能干細(xì)胞的分離與鑒定不論對(duì)于基礎(chǔ)研究還是臨床應(yīng)用都具有非常重要的意義和不可低估的價(jià)值。 本文的第一部分,是從小鼠胚胎成纖維細(xì)胞中成功分離出一群原始的Sca-1~((+))CD117~((-))Lin~((-))多潛能干細(xì)胞,對(duì)于其生物學(xué)特性進(jìn)行了全面而細(xì)致的研究。發(fā)現(xiàn)該細(xì)胞群表達(dá)胚胎干細(xì)胞相關(guān)的轉(zhuǎn)錄因子,具有朝三胚層細(xì)胞分化的能力,并且擁有免疫調(diào)節(jié)性能。在體實(shí)驗(yàn)表明該細(xì)胞群能夠延長異基因來源的皮膚移植物的存活時(shí)間,皮膚燒傷實(shí)驗(yàn)證明其能夠顯著促進(jìn)皮膚損傷的愈合。我們的實(shí)驗(yàn)為胚胎后亞全能干細(xì)胞在體內(nèi)的存在提供了進(jìn)一步的支持證據(jù),豐富了成體干細(xì)胞可塑性的基本理論。第二部分目的是考察Sca-1~((+))CD117~((-))Lin~((-))多潛能干細(xì)胞是否能夠在體外朝造血細(xì)胞分化。使用合適的誘導(dǎo)條件,我們誘導(dǎo)Sca-1~((+))CD117~((-))Lin~((-))分化為具有免疫調(diào)節(jié)功能的樹突狀細(xì)胞,對(duì)于分化而來的樹突狀細(xì)胞我們?cè)敿?xì)的考察了其生物學(xué)特性。進(jìn)一步發(fā)現(xiàn),這些樹突狀細(xì)胞通過Notch-Jagged信號(hào)通路的調(diào)節(jié)而發(fā)揮免疫抑制的效應(yīng)。提示這類誘導(dǎo)的樹突狀細(xì)胞有可能在臨床上具有治療免疫相關(guān)性疾病的前景。
[Abstract]:More and more studies have found that adult stem cells have multi lineage differentiation and the characteristics of the adult stem cell plasticity, and the causes and mechanisms of plasticity, more scholars think there might be primitive pluripotent stem cells in adult tissues, we named pluripotent stem cell embryos for pluripotent stem cell isolation and identification is very important for both basic research and clinical application significance and the value cannot be underestimated.
The first part of this paper, from mouse embryonic fibroblasts isolated from a group of original Sca-1~ ((+)) CD117~ ((-)) Lin~ ((-)) are multipotent stem cells, for its biological characteristics were studied carefully. The cells expressed in embryonic stem cell transcription factor the ability of cell differentiation toward the three germ layers, and have immunomodulatory properties. In vivo experiments showed that the skin cells can prolong allogeneic sources of allograft survival, skin burn experiments prove that it can significantly promote the healing of skin injury. Our experiments for pluripotent stem cells provide supporting evidence further in vivo embryo, enrich the basic theory of adult stem cell plasticity. The second part is to study the Sca-1~ ((+)) CD117~ ((-)) Lin~ ((-)) are multipotent stem cells in vitro in hematopoietic cells can divide . use the induction of appropriate conditions, we induced Sca-1~ ((+)) CD117~ ((-)) Lin~ ((-)) differentiate into immune function of dendritic cells, and dendritic cells for differentiation to our detailed study of its biological characteristics. Further found that these dendritic cells by regulating Notch-Jagged signaling and play effects of immune suppression. Suggesting that these dendritic cells induced by treatment of immune related diseases may have the prospect in clinic.
