骨髓基質(zhì)干細(xì)胞定向分化為神經(jīng)細(xì)胞及其功能特征的研究

【摘要】:目的探討骨髓基質(zhì)干細(xì)胞(BMSCs)定向分化為神經(jīng)細(xì)胞的潛能及分化后細(xì)胞的功能特征。 方法體外分離、擴(kuò)增、純化鼠BMSCs,取第6代細(xì)胞接種并隨機(jī)分為4組:實(shí)驗(yàn)組A組加預(yù)誘導(dǎo)液(EGF+bFGF),其余各組在預(yù)誘導(dǎo)后加定向誘導(dǎo)劑(SHH+RA)時選擇是否保留預(yù)誘導(dǎo)液分為B組(保留)和C組(去除),對照組D組不做任何干預(yù)。采用細(xì)胞免疫熒光化學(xué)法檢測分化后神經(jīng)樣細(xì)胞的形態(tài)和功能標(biāo)志。 結(jié)果BMSCs在誘導(dǎo)劑的特定組合和時序的誘導(dǎo)下,各組均出現(xiàn)胞體收縮和突起伸出,呈現(xiàn)神經(jīng)元細(xì)胞樣改變,B組神經(jīng)細(xì)胞的形態(tài)標(biāo)志MAP2(72.536±1.018)%、GFAP(21.536±1.018)%明顯優(yōu)于其他各組,且功能標(biāo)志ChAT(45.112±3.044)%、VAT(28.223±1.716)%只有B組表達(dá)。 結(jié)論BMSCs在體外能定向誘導(dǎo)為具有一定功能的神經(jīng)細(xì)胞,為脊髓損傷后細(xì)胞移植治療奠定了基礎(chǔ)。
[Abstract]:Objective to investigate the potential of bone marrow stromal cells (BMSCs) to differentiate into neural cells and the functional characteristics of differentiated cells.Methods isolated and amplified in vitro,BMSCs were purified, and the 6th passage cells were inoculated and randomly divided into 4 groups: group A in experimental group with EGF bFGFA, and group A in group A with directional inducer SHH RAA after preinduction, and the other groups were divided into group B (retention) and group B (retention) when adding directional inducer (SHH RAA) after preinduction.Group D did not intervene.The morphological and functional markers of differentiated nerve-like cells were detected by immunofluorescence assay.Results under the induction of specific combination of inducers and time series, BMSCs cells contracted and protruded in all groups, and the neuronal cell like changes were observed in group B (MAP2(72.536 鹵1.018%), which was much better than that in other groups (21. 536 鹵1. 018%), and the expression of neuronal cells in group B was significantly higher than that in group B (21. 536 鹵1. 018%).The functional marker ChAT(45.112 鹵3.044 was only expressed in group B (28.223 鹵1.716%).Conclusion BMSCs can be induced into neural cells with certain function in vitro, which lays a foundation for cell transplantation after spinal cord injury.
