【摘要】：目的檢測Ⅱ型糖尿病(T2DM)C57BL/6小鼠外周血及脾臟中白細胞介素-17(IL-17)、β轉(zhuǎn)化生長因子(TGF-β)、白細胞介素-6(IL-6)的變化。方法 C57BL/6小鼠糖尿病造模成功后,于d 3、d 9、d 15、d 21取血及脾臟,用ELISA方法檢測小鼠外周血中IL-17、TGF-β、IL-6水平變化及用RT-PCR檢測小鼠脾臟IL-17、TGF-β、IL-6 m RNA的表達情況。結(jié)果小鼠造模成功后的d 3、d 9、d 15、d 21,外周血IL-17水平在d 3、d 9高于對照組(P0.05);TGF-β水平在d 3高于對照組(P0.05),隨時間的增長呈下降趨勢;IL-6水平呈上升趨勢,在d 9、d15高于對照組(P0.05)。脾臟中IL-17在d 3、d 9高于對照組(P0.05);TGF-β表達呈下降趨勢,在d 3高于對照組(P0.05);IL-6表達呈上升趨勢,在d 9、d 15、d 21高于對照組(P0.05)。結(jié)論Ⅱ型糖尿病C57BL/6小鼠模型中IL-17、TGF-β的表達在造模早期呈升高趨勢,IL-6的表達高于對照組,并在d 15達到高峰。該研究結(jié)果為Th17亞群在T2DM發(fā)生發(fā)展進程中發(fā)揮的免疫學作用提供了動態(tài)的理論數(shù)據(jù),并為臨床免疫治療提供理論基礎(chǔ)。
[Abstract]:Objective to detect the changes of interleukin-17 (IL-17), 尾 transforming growth factor TGF- 尾 (TGF- 尾) and interleukin-6 (IL-6) in peripheral blood and spleen of T2DMN C57BL / 6 mice. The level of IL-6 in peripheral blood of mice was detected by ELISA method, and the expression of IL-6 m RNA in spleen of mice was detected by RT-PCR. Results the level of IL-17 in peripheral blood was higher than that in control group P0.05TGF- 尾 on day 3, day 9, day 15 and day 21 after successful establishment of mice model. (3) the level of IL-6 was higher than that of the control group (P 0.05), and the level of IL-6 increased with the increase of time. The expression of TGF- 尾 in spleen on d 3d 9 was higher than that in control group (P 0.05), and the expression of TGF- 尾 in spleen was higher than that in control group on d 3. The expression of TGF- 尾 in spleen was higher than that in control group (P 0.05), and the expression of TGF- 尾 in spleen was higher than that in control group on d 3.
