人胚胎三叉神經(jīng)節(jié)細(xì)胞形態(tài)及局部定位

【摘要】: 目的:觀察人胚胎三叉神經(jīng)節(jié)的形態(tài)結(jié)構(gòu)及研究人胚胎發(fā)出眼神經(jīng)、上頜神經(jīng)及下頜神經(jīng)的神經(jīng)元在三叉神經(jīng)節(jié)內(nèi)的定位。方法:收集胎齡為20周以上的胎兒9例,灌注后進行分組:形態(tài)學(xué)觀察組4例,胎齡為24-28周,左側(cè)三叉神經(jīng)節(jié)進行蘇木素-伊紅染色,右側(cè)進行醋酸雙氧鈾-檸檬酸鉛染色;其余5例胎齡為20-26周標(biāo)本為DiI染色晶體植入組,3例左側(cè)眼神經(jīng)植入、右側(cè)上頜神經(jīng)植入,1例雙側(cè)下頜神經(jīng)植入,1例左側(cè)下頜神經(jīng)、右側(cè)三叉神經(jīng)根植入,最后通過光鏡、電鏡及熒光顯微鏡等方法進行形態(tài)學(xué)觀察,拍照后在Adobe Photoshop(7.0,中文版)處理,分析結(jié)果。結(jié)果:①光鏡下節(jié)細(xì)胞的胞質(zhì)內(nèi)可見粗大的尼氏顆粒,細(xì)胞核近圓形,呈空泡狀,核仁基本居中;②電鏡下節(jié)細(xì)胞的胞質(zhì)內(nèi)細(xì)胞器豐富,可見大量線粒體、發(fā)育較好的高爾基復(fù)合體、粗面內(nèi)質(zhì)網(wǎng)及游離核糖體,核膜凹凸不平,核孔多;③熒光顯微鏡下三叉神經(jīng)節(jié)內(nèi)可見到DiI標(biāo)記的節(jié)細(xì)胞和神經(jīng)纖維,節(jié)細(xì)胞的胞體呈圓形或橢圓形,大小不一,可見連于胞體的突起,在神經(jīng)節(jié)內(nèi)還可見到雙極神經(jīng)元;④人胚胎三叉神經(jīng)節(jié)是沿內(nèi)外方向分布的細(xì)胞團,發(fā)出眼神經(jīng)的神經(jīng)元位于神經(jīng)節(jié)的前內(nèi)側(cè),下頜神經(jīng)的神經(jīng)元位于神經(jīng)節(jié)的后外側(cè),上頜神經(jīng)的神經(jīng)元位于兩者之間。結(jié)論:胎齡24-28周人胚胎三叉神經(jīng)元顯示為成熟細(xì)胞特征;人胚胎發(fā)出眼神經(jīng)、上頜神經(jīng)及下頜神經(jīng)的神經(jīng)元在三叉神經(jīng)節(jié)內(nèi)以從內(nèi)向外的局部定位方式排列。
[Abstract]:Objective: to observe the morphological structure of the trigeminal ganglion of human embryo and to study the localization of the neurons of the human embryonic emitting ophthalmic nerve, maxillary nerve and mandibular nerve in the trigeminal ganglion. Methods: nine fetuses over 20 weeks of gestational age were collected. After perfusion, there were 4 cases in the morphological observation group with gestational age of 24-28 weeks. The left trigeminal ganglion was stained with hematoxylin eosin, and the right side was stained with dioxy-uranium-citrate lead acetate. The other 5 cases of 20-26 weeks of gestational age were implanted with DiI staining lens. The left eye nerve was implanted in 3 cases, the bilateral mandibular nerve was implanted in 1 case and the right trigeminal nerve root was implanted in the left mandibular nerve in 1 case, and the root of the right trigeminal nerve was implanted with light microscope. Electron microscope and fluorescence microscope were used to observe the morphology of the ganglion cells. The results were analyzed by Adobe Photoshop 7.0. results under the light microscope of 1: 1, the cytoplasm of the ganglion cells could be seen in the thick Nissl granules, the nucleus was nearly round, and the cells were vacuolated. The cytoplasmic organelles of the nucleus were abundant in the inferior ganglion cells under electron microscope. A large number of mitochondria, well developed Golgi complex, rough endoplasmic reticulum and free ribosome were observed, and the ribosome was uneven. DiI labeled ganglion cells and nerve fibers could be seen in the trigeminal ganglion under the fluorescence microscope of the nuclear foramen. The ganglion cells were round or oval in size and different in size, and the processes attached to the cell bodies could be seen. In the ganglion, the trigeminal ganglion of 4 human embryos is a cell cluster distributed in the inner and outer directions. The neurons of the ophthalmic nerve are located on the anterior medial side of the ganglion, and the neurons of the mandibular nerve are located on the lateral side of the ganglion. Conclusion: the trigeminal neurons of human embryo at 24-28 weeks of gestational age show the characteristics of mature cells. The neurons of the maxillary nerve and the mandibular nerve are arranged in the trigeminal ganglion in an inward and outward local location.
