【摘要】：目的：本實驗通過結(jié)扎小鼠一側(cè)或雙側(cè)輸精管,造成輸精管梗阻,精子輸出受阻,光鏡觀察睪丸、附睪的變化,檢測小鼠外周血CD4+CD25+調(diào)節(jié)性T細(xì)胞,探討輸精管梗阻小鼠睪丸的形態(tài)學(xué)變化以及梗阻對CD4+CD25+調(diào)節(jié)性T細(xì)胞的影響。方法：選用健康成年雄性BALB/c小鼠,隨機(jī)分為3組,每組30只小鼠：對照組(A組,麻醉后僅進(jìn)行陰囊切開后縫合),單側(cè)輸精管結(jié)扎組(B組,麻醉后結(jié)扎左側(cè)輸精管),雙側(cè)輸精管結(jié)扎組(C組,麻醉后兩側(cè)輸精管均結(jié)扎)。分別手術(shù)后10周、20周、26周(每個時間點各組取10只小鼠)應(yīng)用光鏡觀察石蠟包埋HE染色后的小鼠睪丸、附睪的組織結(jié)構(gòu),流式細(xì)胞術(shù)檢測外周血CD4+CD25+CD127-調(diào)節(jié)性T細(xì)胞百分比。結(jié)果：1、HE染色觀察：輸精管結(jié)扎小鼠睪丸形態(tài)學(xué)變化主要表現(xiàn)為結(jié)扎10周組生精小管結(jié)構(gòu)不清,生精細(xì)胞有減少。2、輸精管結(jié)扎一段時間后,實驗小鼠外周血CD4+CD25+CD127-調(diào)節(jié)性T細(xì)胞百分比較對照組明顯增高。結(jié)論：隨著輸精管梗阻時間的延長,機(jī)體對精子產(chǎn)生了免疫耐受,CD4+CD25+CD127-調(diào)節(jié)性T細(xì)胞顯著增加,參與誘導(dǎo)免疫耐受的產(chǎn)生。
[Abstract]:Objective: to observe the changes of testis and epididymis by ligating one or both vas deferens of mice, resulting in obstruction of vas deferens and blocked sperm output, and to detect the regulatory T cells of CD4 CD25 in peripheral blood of mice. To investigate the morphological changes of testis and the effect of obstruction on CD4 CD25 regulatory T cells in mice with vasectomy. Methods: healthy adult male BALB/c mice were randomly divided into 3 groups, 30 mice in each group: control group A; After anesthesia, only scrotal incision and suture were performed, unilateral vasectomy group (group B), left vasectomy group (group C) and bilateral vasectomy group (group C). After anesthesia, both vas deferens were ligated. The tissues of the testis and epididymis were observed by light microscope at 10 weeks and 20 weeks after operation (10 mice in each group were taken at each time point), and the tissues of the testis and epididymis were observed by light microscope after paraffin-embedded HE staining. The percentage of CD4 CD25 CD127- regulatory T cells in peripheral blood was detected by flow cytometry. Results the morphological changes of testis in vas deferens ligated mice were characterized by unclear structure of seminiferous tubules after 10 weeks of ligation. The percentage of CD4 CD25 CD127- regulatory T cells in peripheral blood of the experimental mice was significantly higher than that of the control group. Conclusion: with the prolongation of the obstructive time of the vas deferens, the percentage of CD127- regulatory T cells in peripheral blood of the experimental mice was significantly higher than that of the control group. Immune tolerance T cells (CD4 CD25 CD127- regulatory T cells) produced by organism to spermatozoa were significantly increased, which was involved in the induction of immune tolerance.
