健康人和HIV感染者非特異性IgG,IgG1-4及gp41特異性IgG1的研究

【摘要】: 研究背景免疫球蛋白G (immunoglobulin G, IgG)有四個(gè)亞型[1]。IgG1-4水平的變化往往意味著機(jī)體免疫力受損或者受到病原微生物的感染[2-5],因此檢測IgG1-4水平可以用來評估HIV感染者免疫水平和疾病進(jìn)程[6]。一些國家和地區(qū)的研究結(jié)果證明,IgGl-4表達(dá)水平在不同人種間存在差異[7];對于HIV感染者的相關(guān)研究顯示,HIV感染者擁有較高的IgG1和IgG3,IgG2低于對照人群,IgG4則無顯著性差異[8]。我國在該領(lǐng)的研究非常有限。 本研究以北京市健康成人和貴州、重慶兩地的HIV感染者為研究對象,開展了健康人IgG、IgG1-4, HIV感染者IgG, IgG1-4和gp41特異性IgG1的研究。該研究對于了解我國健康人和HIV感染者免疫水平、特異性免疫反應(yīng),發(fā)展相關(guān)的免疫學(xué)檢測方法具有重要意義。 研究目的1、研究測定北京市健康成人IgG. IgG1-4水平,為其他研究提供基礎(chǔ)資料。2、研究測定HIV感染者IgG、IgG1-4水平,并對不同感染途徑人群的結(jié)果進(jìn)行比較研究,探討將其作為免疫學(xué)評價(jià)的指標(biāo)。3、研究HIV感染者gp41特異性IgG1的產(chǎn)生和變化,探討作為潛在新近感染檢測指標(biāo)的可能性。 材料和方法收集健康人群血漿315份;HIV陽性IDU人群血漿222份;HIV陽性MSM人群血漿115份;HIV陽性隊(duì)列樣本80份。用夾心ELISA法定量檢測血漿中IgG水平;用競爭ELISA方法定量檢測血漿中IgG1-4水平;用間接ELISA法定量檢測HIV陽性人群血漿中g(shù)p41特異性IgG1。對實(shí)驗(yàn)結(jié)果采用非參數(shù)秩和檢驗(yàn)進(jìn)行統(tǒng)計(jì)分析[9]。 研究結(jié)果1、北京市健康成人IgG, IgG1-4水平分別為:4.83mg/ml、7.84 mg/ml、3.55 mg/ml、0.34 mg/ml和0.23 mg/ml; IgG、IgG4在性別間有差異;IgGl-3有隨著年齡增長而下降的趨勢。2、HIV陽性人群IgG1和IgG3均有隨著CD4細(xì)胞計(jì)數(shù)的升高而增設(shè)的趨勢;HIV陽性人群IgG與健康成人相比無差異;HIV陽性IDU人群IgG2與CD4細(xì)胞計(jì)數(shù)之間的關(guān)系與MSM人群截然相反;HIV陽性MSM人群IgG4與健康成人相比無統(tǒng)計(jì)學(xué)差異。HIV陽性IDU、MSM人群IgG IgG1-4存在統(tǒng)計(jì)學(xué)差異。 3、HIV陽性人群gp41特異性IgG1隨著CD4細(xì)胞計(jì)數(shù)的下降有降低的趨勢,但變化趨勢不明顯。HIV陽性IDU、MSM人群之間gp41特異性IgG1有統(tǒng)計(jì)學(xué)差異。4、gp41特異性IgG1隨著陽轉(zhuǎn)時(shí)間的延長有升高的趨勢,但升高的幅度不明顯;168天以內(nèi)感染者與168以上感染者gp41特異性IgG1濃度無統(tǒng)計(jì)學(xué)差異。 研究結(jié)論1、北京市健康成人IgG、IgG1-4水平分別為:4.83mg/ml,7.84 mg/ml、3.55 mg/ml、0.34 mg/ml和0.23 mg/ml。2、可以嘗試用IgG1和Ig3做為評估HIV感染者免疫狀況或疾病進(jìn)程的指標(biāo);IgG、IgG2和IgG4不宜作為評價(jià)HIV感染者免疫狀態(tài)的指標(biāo)。不同傳播途徑對IgG、IgG1-4的表達(dá)水平可能造成影響。3、不宜用gp41特異性IgG1作為評價(jià)HIV感染者免疫狀況或病痛進(jìn)程的指標(biāo)。不同傳播途徑對gp41特異性IgG1可能造成影響。4、不宜用gp41特異性IgG1作為識別新近和既往長期感染的檢測指標(biāo)。
[Abstract]:Protein research background of immunoglobulin G (immunoglobulin G IgG) change four subtype [1].IgG1-4 levels often mean immunity damaged or infected [2-5] of pathogenic microorganisms, so to detect the level of IgG1-4 can be used to study the evaluation of HIV infection immunity and disease progression of [6]. in some countries and regions the results show that the expression level of IgGl-4 in the different people are different [7] species; for the research of HIV infection, HIV infection had higher IgG1 and IgG3, IgG2 lower than the control group IgG4, there was no significant difference of [8]. in our country is very limited in the research area.
In this study, healthy adults in Beijing city and Guizhou, Chongqing two HIV infection as the research object, the health of human IgG, IgG1-4, HIV and IgG1-4 of IgG infection, gp41 specific IgG1. The study to understand the immune level of healthy people in China and HIV infection, specific immune response is important the significance of the development of immunological detection methods.
Objective: 1, determination of IgG1-4 level of healthy adults in Beijing city IgG..2, to provide basic data for other studies, the researchers measured IgG HIV infection, IgG1-4 level, and the population of different routes of infection are compared with the results of the study, explore the immunological evaluation index of.3 as the emergence and change of gp41 specific IgG1 research to explore the possibility of HIV infection, as a potential new infections detection index.
Materials and methods: We collected samples of 315 healthy population; HIV positive among IDU samples of 222 HIV positive MSM patients; 115 plasma samples; HIV positive cohort 80 samples by IgG. The level of quantitative sandwich ELISA detection in plasma; by competitive ELISA method for quantitative detection of plasma IgG1-4 levels in plasma; ELISA method for quantitative detection of HIV positive people gp41 specific IgG1. on experimental results using the nonparametric Wilcoxon test was used for statistical analysis [9].
The results of 1 healthy adults in Beijing City, IgG, IgG1-4 levels were 4.83mg/ml, 7.84 mg/ml, 3.55 mg/ml, 0.34 mg/ml and 0.23 mg/ml; IgG IgG4, there are differences in the different sex; IgGl-3 has decreased with the age growth trend of.2, IgG1 and IgG3 were HIV positive patients with elevated CD4 cell count and additional the trend of HIV; IgG positive people with healthy adults compared with no difference between IgG2 and MSM; the relationship between population HIV positive IDU patients with CD4 cell counts in the opposite; IgG4 HIV positive MSM group no significant difference in the.HIV positive IDU compared with healthy adults, there were significant differences among MSM IgG IgG1-4.
