應(yīng)用微列陣技術(shù)快速鑒定常見致病性分枝桿菌

【摘要】: 【目的】利用微列陣技術(shù)建立一種簡便、快速、靈敏、特異的分支桿菌菌種鑒定方法,為臨床醫(yī)師正確診斷提供依據(jù)。 【方法】以DNA測序法為對照,通過微列陣技術(shù)分析20種分枝桿菌標(biāo)準(zhǔn)株及486株分枝桿菌臨床分離株。 【結(jié)果】應(yīng)用微列陣技術(shù)分析,20種分枝桿菌標(biāo)準(zhǔn)株鑒定結(jié)果的特異性為100%,486株分枝桿菌臨床分離株中,顯示358株臨床分離株與分枝桿菌屬探針I(yè)C和結(jié)核分枝桿菌復(fù)合群探針雜交陽性,鑒定為結(jié)核分枝桿菌復(fù)合群(MTC)。128株為非結(jié)核分枝桿菌(NTM),其中47株為龜分枝桿菌龜亞種和膿腫亞種,32株為胞內(nèi)分枝桿菌,15株為鳥分枝桿菌株,11株為堪薩斯分枝桿菌,4株為偶然分枝桿菌,4株為恥垢分枝桿菌,4株為;驖兎种U菌,3株為蘇加爾分枝桿菌或瑪爾摩分枝桿菌,2株為戈登分枝桿菌,2株為土分枝桿菌,1株為瘰疬分枝桿菌,1株為不產(chǎn)色分枝桿菌,2株只與分枝桿菌屬探針雜交,經(jīng)測序顯示1株為副瘰疬分枝桿菌,1株為莫娜分枝桿菌,芯片上無鑒定這2個菌種的探針。 【結(jié)論】微列陣技術(shù)是一種準(zhǔn)確、高效的快速鑒定分枝桿菌菌種的分子診斷技術(shù)。
[Abstract]:[objective] to establish a simple, rapid, sensitive and specific method for the identification of mycobacteria by using microarray technique. [methods] 20 standard mycobacterium strains and 486 clinical isolates of Mycobacterium were analyzed by microarray technique using DNA sequencing method as control. [results] the specificity of the identification of 20 standard strains of Mycobacterium by microarray technique was 100 and 486. The results showed that 358 clinical isolates were positive for hybridization with IC and Mycobacterium tuberculosis complex probes. Identified as Mycobacterium tuberculosis complex MTCN. 128 strains as non-tuberculous mycobacteria NTMN, of which 47 strains are Mycobacterium tortoise subspecies and 32 strains of abscess subspecies, 15 strains of Mycobacterium endogenes, 15 strains of Mycobacterium avium, 11 strains of Mycobacterium Kansan, 11 strains of Mycobacterium Kansas. 4 strains of Mycobacterium fortuitum, 4 strains of Mycobacterium smearus, 4 strains of Mycobacterium ulcerans, 3 strains of Mycobacterium thuringiensis or 2 strains of Mycobacterium malmo, 2 strains of Mycobacterium Gordon, 2 strains of Mycobacterium tumefaciens, 1 strain of Mycobacterium scrofula, 1 strain of Mycobacterium scrofulla. One strain of Mycobacterium, one of which is a non-chromogenic mycobacterium, only hybridized with a probe of the genus Mycobacterium. Sequencing showed that one strain was Mycobacterium parascrofula and one strain was Mycobacterium mona. There was no probe on the microarray to identify the two strains. [conclusion] Microarray technique is an accurate and efficient molecular diagnostic technique for the identification of Mycobacterium species.
