分泌表達(dá)結(jié)核分枝桿菌Ag85B蛋白的重組卡介苗的構(gòu)建和免疫學(xué)特性的初步研究

【摘要】: 結(jié)核病是由結(jié)核分枝桿菌引起的以呼吸道傳播為主的慢性傳染病,結(jié)核病被WHO稱為唯一的一種全球性緊迫性疾病。據(jù)WHO報(bào)道,全世界目前約有1?3人口潛伏感染結(jié)核桿菌,2005年新增病例約為880萬,死亡160萬。貧窮、人口增加、流動(dòng)人口增多、耐藥菌株及艾滋病流行等使結(jié)核病雪上加霜。我國結(jié)核患者數(shù)量居世界第二位,我國現(xiàn)有結(jié)核感染者4億人。 卡介苗是目前世界上唯一可以應(yīng)用的預(yù)防結(jié)核病的疫苗。但卡介苗在不同地區(qū)不同人群中存在著差異?ń槊缈梢杂行ьA(yù)防兒童型結(jié)核感染,但不能預(yù)防成人型肺結(jié)核,其預(yù)防作用從0-80%不等。由于BCG預(yù)防結(jié)核病的不足,以及當(dāng)前TB流行的嚴(yán)重性,因此研究新的疫苗用于結(jié)核的預(yù)防已成為當(dāng)前國內(nèi)外研究的熱點(diǎn)。本研究主要構(gòu)建了表達(dá)Ag85B的重組卡介苗,以彌補(bǔ)傳統(tǒng)卡介苗在預(yù)防結(jié)核病的不足。用分泌表達(dá)Ag85B蛋白的rBCG和普通BCG分別經(jīng)皮下免疫BALB/c小鼠,評價(jià)各組免疫小鼠的特異性體液免疫和細(xì)胞免疫水平。 實(shí)驗(yàn)結(jié)果表明分泌表達(dá)Ag85B蛋白的rBCG與普通BCG相比可以大量分泌抗原Ag85B;長時(shí)間免疫后的檢測結(jié)果表明rBCG比BCG刺激較強(qiáng)的體液免疫應(yīng)答,但在細(xì)胞免疫反應(yīng)上不是很強(qiáng)。因此,rBCG的免疫保護(hù)作用還有待于進(jìn)一步研究。
[Abstract]:Tuberculosis is a chronic infectious disease mainly transmitted by respiratory tract caused by Mycobacterium tuberculosis. Tuberculosis is regarded as the only global urgent disease by WHO. According to WHO, there are about 1? In 2005, the number of new cases was about 8.8 million and the death rate was 1.6 million. Poverty, population increase, floating population increasing, drug-resistant strains and the prevalence of HIV / AIDS made TB worse. The number of TB patients in China ranked second in the world. There are 400 million people infected with tuberculosis in China. BCG vaccine is the only vaccine available in the world to prevent tuberculosis. But BCG vaccine varies from region to region. BCG vaccine can effectively prevent tuberculosis infection in children, but it can not prevent tuberculosis in adults. The preventive effects range from 0 to 80%. Because of the insufficient prevention of TB by BCG and the severity of the current TB epidemic, Therefore, the research on the application of new vaccines to the prevention of tuberculosis has become a hot topic at home and abroad. In this study, recombinant BCG vaccine expressing Ag85B was constructed. In order to make up for the deficiency of traditional BCG vaccine in preventing tuberculosis, BALB/c mice were immunized subcutaneously with rBCG and BCG which secreted Ag85B protein to evaluate the specific humoral immunity and cellular immunity of each group of immunized mice. The results showed that rBCG secreting Ag85B protein could secrete Ag85B in a large amount compared with ordinary BCG, and the results of long time immunization showed that rBCG stimulated a stronger humoral immune response than BCG. But the cellular immune response is not very strong, so the protective effect of rBCG remains to be further studied.
