轉(zhuǎn)染HCN2基因的間充質(zhì)干細胞對兔病竇模型的影響

【摘要】:目的 尋找一種能長期穩(wěn)定的兔病竇模型的制作方法,并檢測轉(zhuǎn)染了HCN2基因的間充質(zhì)干細胞對兔病竇模型的影響。 方法 日本大耳白兔隨機分組,分別用射頻消融法和甲醛濕敷法制作逸病竇模型。兔麻醉后經(jīng)胸骨右緣開胸,大頭電極于上腔靜脈和右心房交接處竇房結(jié)區(qū)域放電,以竇性心率下降30-50%或出現(xiàn)交界性逸搏心律為消融成功標志,并且甲醛濕敷法做對照。觀察兩種方法后兔的心率變化,并分別測其固有心率。體外培養(yǎng)間充質(zhì)干細胞,提取含HCN2和GFP的質(zhì)粒,并將質(zhì)粒轉(zhuǎn)染到間充質(zhì)干細胞上,注射入病竇兔右心房,對照組用生理鹽水注射,觀察轉(zhuǎn)染了HCN2基因的間充質(zhì)干細胞對兔病竇模型的影響。 結(jié)果 射頻消融法制作的兔病竇模型較甲醛濕敷法組心率及固有心率均明顯減慢,甲醛濕敷法組較與正常組比較亦沒有明顯差異。注射了含HCN2基因間充質(zhì)干細胞的兔心率與對照組相比,沒有明顯差別。 結(jié)論 與常用的甲醛消融法相比,射頻消融法制作兔病竇模型是一種更優(yōu)化的方法。兔右心房不是構(gòu)建生物起搏器的適宜注射位點。
[Abstract]:Purpose. To find a long-term stable rabbit sinus model and detect the effect of mesenchymal stem cells (MSCs) transfected with HCN2 gene on rabbit sick sinus model. Method. Japanese large ear white rabbits were randomly divided into two groups: radiofrequency ablation method and formaldehyde wet compress method, respectively. After anesthesia, the right sternum was opened and the large head electrode was discharges in the sinus node area at the junction of superior vena cava and right atrium. The sinus heart rate decreased by 30-50% or the borderline escape rhythm was used as the successful sign of ablation, and the formaldehyde wet compress method was used as the control. The changes of heart rate were observed and the intrinsic heart rate was measured respectively. The mesenchymal stem cells were cultured in vitro. The plasmids containing HCN2 and GFP were extracted and transfected into the mesenchymal stem cells (MSCs) and injected into the right atrium of the diseased sinus rabbits. The control group was injected with normal saline to observe the effect of the mesenchymal stem cells (MSCs) transfected with HCN2 gene on the rabbit model of diseased sinus. Results. The heart rate and intrinsic heart rate of the rabbit sinus model made by radiofrequency ablation were significantly slower than those of the wet compress group. The heart rate of rabbits injected with HCN2 gene mesenchymal stem cells was not significantly different from that of the control group. Conclusion. Compared with formaldehyde ablation method, radiofrequency ablation is a better method for the establishment of rabbit sick sinus model, and the right atrium is not a suitable injection site for the construction of biological pacemakers.
