【摘要】：目的探討不同地理分布的恒河猴亞種在結(jié)核分枝桿菌感染實驗中的反應(yīng)差異,為在結(jié)核疫苗評價中選擇合適的動物模型提供依據(jù)。方法通過纖維支氣管鏡定量感染湖北、廣西恒河猴,建立急性感染結(jié)核病模型。在不同的時間點檢測血液、血生化(包括C反應(yīng)蛋白)、血沉等指標,并對組織細菌載量、肺組織和淋巴結(jié)病理切片進行對比分析。結(jié)果湖北恒河猴的血液學(xué)指標在感染期間均呈下降趨勢,在第16、20周(感染后期)均低于廣西恒河猴,但血沉值在12、16、20周呈上升趨勢,均高于廣西恒河猴。兩組恒河猴血液中性粒細胞在第12周(湖北猴1.1×1010/L,廣西猴4.7×109/L)、淋巴細胞第4周(湖北猴1.7×109/L,廣西猴6.8×109/L)和嗜堿性粒細胞在感染前(湖北猴4.3×107/L,廣西猴2.0×108/L)差異有統(tǒng)計學(xué)意義(P0.01),血小板在16周(湖北猴3.8×1011/L,廣西猴4.5×109/L)、20周(湖北猴4.1×1011/L,廣西猴4.9×109/L)差異有統(tǒng)計學(xué)意義(P0.01)。湖北恒河猴左肺細菌載量(4.52lgCFU/g)低于廣西恒河猴(6.30lgCFU/g)(P0.05)。與廣西恒河猴相比較,湖北恒河猴左、中、上肺有明顯結(jié)核結(jié)節(jié)。結(jié)論在MTB的急性感染實驗中,湖北和廣西恒河猴的血液學(xué)指標、部分組織的細菌載量和病理改變均存在差異。
[Abstract]:Objective to explore the difference of the response of different geographical distribution of rhesus monkey subspecies in Mycobacterium tuberculosis infection test, and to provide the basis for selecting suitable animal model in the evaluation of tuberculosis vaccine. In Guangxi rhesus monkey, an acute infection tuberculosis model was established. Blood, blood biochemistry (including C-reactive protein, erythrocyte sedimentation rate, etc.) and bacterial load in tissues were detected at different time points. Results the hematological indexes of Hubei rhesus monkey showed a downward trend during infection, and were lower than those of Guangxi rhesus monkey at the 20th week of infection (late infection), but the ESR showed an increasing trend at 1216 ~ 20 weeks. The blood neutrophils of the two groups were higher than those of the Guangxi rhesus monkeys at the 12th week (1.1 脳 10 10 / L in Hubei, 4.7 脳 10 9 / L in Guangxi, 1.7 脳 10 9 / L in Hubei, 6.8 脳 10 9 / L in Guangxi) and basophil before infection (4.3 脳 10 7 / L in Hubei monkey, 2.0 脳 10 8 / L in Guangxi monkey). There was a statistically significant difference in platelet levels at 16 weeks (3.8 脳 1011 / L in Hubei monkey, 4.5 脳 109 / L / L in Guangxi monkey, 4.1 脳 1011 / L in Hubei monkey and 4.9 脳 109 / L in Guangxi monkey). The bacterial load in left lung of Hubei rhesus monkey was lower than that of Guangxi rhesus monkey (6.30lg CFU P 0.05g). There are obvious tuberculous nodules in the left, middle and upper lungs of Hubei rhesus monkey. Conclusion in the acute infection experiment of MTB, the hematological indexes, bacterial load and pathological changes in some tissues of Hubei and Guangxi rhesus monkeys are different.
