Decorin對人僵直膝關(guān)節(jié)滑膜囊成纖維樣滑膜細(xì)胞增殖及蛋白合成的抑制作用

【摘要】:[目的]探討重組核心蛋白多糖(decorin)對培養(yǎng)的人僵直膝關(guān)節(jié)滑膜成纖維樣滑膜細(xì)胞(fibroblast-like synoviocytes,FLS)的增殖和Ⅰ型膠原蛋白及mRNA合成的影響,為Decorin治療人膝關(guān)節(jié)僵直提供理論依據(jù)。[方法]分離培養(yǎng)人僵直膝關(guān)節(jié)滑膜FLS,分別加入不同質(zhì)量濃度(10μg/ml,0.1μg/ml)的Decorin;培養(yǎng)24、48、72h后用二甲基噻唑二苯基四唑溴鹽(MTT)法測定FLS的增殖速度;用流式細(xì)胞儀檢測FLS的周期和凋亡;用RTPCR法檢測FLS合成Ⅰ型前膠原蛋白(PcⅠ)mRNA的含量,westem Blot檢測Ⅰ型膠原蛋白的合成量,并與對照組比較。[結(jié)果]10μg/ml的Decorin可有效抑制滑膜FLS的增殖(P0.05),明顯增加FLS處于G0/G1期的百分比(P0.05),并通過下調(diào)PcⅠmRNA的表達(dá),在轉(zhuǎn)錄水平上抑制PcⅠ的合成;實驗組和對照組均未檢測到終末期凋亡細(xì)胞。[結(jié)論]Decorin可抑制人僵直膝關(guān)節(jié)滑膜FLS的增殖及PcⅠmRNA的表達(dá)和Ⅰ型膠原蛋白合成,在防治膝關(guān)節(jié)僵直中起一定的作用。
