輸注供體未成熟樹突狀細胞對大鼠皮膚移植的影響

【摘要】: 目的 建立穩(wěn)定的大鼠皮膚移植排斥反應(yīng)模型,探討輸注供體未成熟樹突狀細胞減輕排斥反應(yīng)和誘導(dǎo)免疫耐受的作用。 方法 通過建立大鼠自體皮膚移植模型,熟練掌握大鼠皮膚移植方法。繼而用改良的方法擴增未成熟樹突狀細胞:rhG-CSF,5μg/kg,皮下注射,連續(xù)7天后,從骨髓分離、提取樹突狀細胞,體外培養(yǎng)3d。實驗分組:自體移植組、對照1組SD→Wistar、對照2組Wistar→Wistar、和imDC實驗組SD→Wistar,配對組合皮膚移植。于皮膚移植術(shù)中經(jīng)陰莖背靜脈輸注供體未成熟樹突狀細胞5×10~6個,動態(tài)觀察術(shù)后移植皮膚的存活時間,不同組別移植物病理改變,探討供體imD減輕大鼠皮膚移植免疫排斥反應(yīng)的作用。 結(jié)果 1、全部自體移植皮片至觀察終點(>2月)仍然存活,移植皮片與周圍組織愈合良好,無缺損或破潰,色澤與周圍組織無明顯區(qū)別。移植物表皮及附屬毛發(fā)、毛囊生長良好。術(shù)后14d鏡下觀察移植物病理切片可見:各組織層次清晰完整,皮膚附屬結(jié)構(gòu)完好,可以看到完整的毛囊結(jié)構(gòu),細胞結(jié)構(gòu)完好,無明顯的炎性細胞浸潤。2.對照1組,移植皮片在14天內(nèi)全部被排斥掉,排斥反應(yīng)發(fā)生早,且嚴重。移植物的平均存活時間僅為11.8±1.6d,最短者于術(shù)后第9d完全排斥。3.對照2組:Wistar→Wistar組,雖然移植物最終全部被排斥,但排斥反應(yīng)發(fā)生晚且較輕,平均生存期為23.1±2.4d。移植物最長存活27d,最短存活19d。術(shù)后14d鏡下移植物病理切片表現(xiàn):移植物組織結(jié)構(gòu)不完整,附屬結(jié)構(gòu)被破壞,未見正常毛囊結(jié)構(gòu),可見部分壞死灶,有大量炎細胞浸潤。于高倍鏡下可見組織細胞核溶解、碎裂。4.實驗組,全部移植物在25d內(nèi)被排斥,存活時間最短14d,平均存活時間為19.3±3.4d,最短14d。術(shù)后14d鏡下移植物病理切片表現(xiàn):移植物組織結(jié)構(gòu)遭破壞,可見壞死灶,有大量炎細胞浸潤。于高倍鏡下可見組織細胞核溶解、碎裂。 結(jié)論 使用供體imDC預(yù)處理受體鼠可以顯著延長移植物的存活時間,減輕排斥反應(yīng)。
[Abstract]:Purpose To establish a stable model of skin allograft rejection in rats and to explore the role of donor immature dendritic cells in reducing rejection and inducing immune tolerance. Method The rat skin autograft model was established to master the method of rat skin transplantation, and then the immature dendritic cells were amplified by 5 渭 g / kg and injected subcutaneously with a modified method. After 7 days, dendritic cells were isolated from bone marrow and cultured for 3 days in vitro. Experimental group: autologous transplantation group, control group 1 SD 鈫扺istar, control group 2 Wistar. 鈫扺istar, and imDC experimental group SD. 鈫扵he donor immature dendritic cells (5 脳 10 ~ 6) were injected through the dorsal penile vein to observe the survival time of the skin graft. Objective: to investigate the role of donor imD in alleviating the immune rejection of skin transplantation in rats. Results 1. All the autografted skin grafts survived until the observed end point (> February). The skin grafts healed well with the surrounding tissues, and there was no defect or rupture. There was no significant difference in color and color between the grafts and surrounding tissues. The skin follicles grew well with the grafts epidermis and hair. The histological sections of the grafts were clear and complete under the microscope 14 days after operation. It can be seen that the complete hair follicle structure, cell structure intact, no obvious inflammatory cell infiltration. 2. In the control group 1, all the skin grafts were rejected within 14 days, the rejection occurred early. The mean survival time of the graft was only 11.8 鹵1.6 days, and the shortest graft was completely rejected on the 9th day after operation. 鈫捍n the Wistar group, although all the grafts were eventually rejected, the rejection occurred later and lighter, with an average survival time of 23.1 鹵2.4 days. The graft survived for the longest 27 days. The shortest survival time was 19 days. The histologic features of the grafts were incomplete, the accessory structure was destroyed, the normal hair follicle structure was not seen, and some necrotic foci were observed at the 14th day after operation. A large number of inflammatory cells infiltrated. Under the high power microscope, the tissue nucleus was dissolved, fragmentation. 4. In the experimental group, all the grafts were rejected within 25 days, the shortest survival time was 14 days. The mean survival time was 19.3 鹵3.4 days and the shortest was 14 days. A large number of inflammatory cells infiltrated. Under the high power microscope, the nucleus of the tissue was dissolved and broken. Conclusion Pretreatment with donor imDC could significantly prolong the survival time of the grafts and alleviate the rejection.
