β1,4-半乳糖基轉(zhuǎn)移酶I與細(xì)胞間黏附分子1在著床調(diào)控中的相關(guān)性探討

【摘要】: 目的:探討著床前子宮內(nèi)膜細(xì)胞β1,4-GalT-I的表達(dá)及調(diào)控β1,4-GalT-I對細(xì)胞間黏附分子1(ICAM-1)表達(dá)的影響,分析β1,4-GalT-I與ICAM一1在胚胎著床過程中的相關(guān)性。 β1,4-半乳糖基轉(zhuǎn)移酶I(β1,4-GalT-I)分布于高爾基體和細(xì)胞膜兩種亞細(xì)胞結(jié)構(gòu)上,位于高爾基體上的β1,4-GalT-I可行使蛋白質(zhì)的糖基化修飾功能;位于細(xì)胞膜上的β1,4-GalT-I則作為一種細(xì)胞黏附分子,參與細(xì)胞黏附、精卵識別、神經(jīng)軸突的生長、腫瘤細(xì)胞遷移等過程。 細(xì)胞間黏附分子1(intercellular adhesion molecule-1,ICAM-1)既是一類重要的黏附分子,也是一種跨膜糖蛋白,它在早期胚泡發(fā)育、子宮內(nèi)膜容受性的建立及胚泡著床過程中有重要作用。 方法:利用人源的子宮內(nèi)膜癌細(xì)胞(RL95-2)模擬著床前子宮內(nèi)膜,人源的絨毛癌細(xì)胞(JAR)模擬胚泡來組成體外著床模型,將β1,4-GalT-I基因過表達(dá)質(zhì)粒(HA-GalT-I)以及空載體質(zhì)粒(pCDNA3.0)轉(zhuǎn)染RL95-2細(xì)胞使其過表達(dá)β1,4-半乳糖基轉(zhuǎn)移酶Ⅰ,通過細(xì)胞熒光檢測確定轉(zhuǎn)染效果,采用RT-PCR、Western-blot等方法檢測調(diào)控β1,4-GalT-I表達(dá)對ICAM-1表達(dá)的影響,檢測JAR細(xì)胞在RL95-2細(xì)胞上的黏附率判斷二者在胚胎著床調(diào)控中的相關(guān)性。 結(jié)果:轉(zhuǎn)染β1,4-GaT-I過表達(dá)質(zhì)粒HA-GalT-I后60小時,RL95-2細(xì)胞中有綠色熒光蛋白的表達(dá),RT-PCR及Western-blot分析結(jié)果顯示:與正常培養(yǎng)組、空質(zhì)粒轉(zhuǎn)染組相比,過表達(dá)HA-GalT-I組β14-GaT-I的表達(dá)量增高,ICAM-I的基因表達(dá)和蛋白合成也隨之增加;過表達(dá)HA-GalT-I后JAR細(xì)胞在RL95-2細(xì)胞上的黏附率(78.5%)也明顯高于正常培養(yǎng)組(65.4%)和空質(zhì)粒轉(zhuǎn)染組(62.5%),(p0.05),說明上調(diào)β1,4-GalT-I會促進(jìn)ICAM-I表達(dá)及提高胚泡著床率。 結(jié)論: (1)調(diào)控β1,4-GalT-I表達(dá)可影響ICAM-1的表達(dá); (2)β1,4-GalT-I可能通過調(diào)節(jié)ICAM-1的表達(dá)參與著床調(diào)控。
[Abstract]:Objective: to investigate the expression of 尾 _ 1N _ 4-GalT-I and its effect on the expression of intercellular adhesion molecule 1 (ICAM-1) in endometrial cells before implantation, and to analyze the effect of 尾 _ 1 on the expression of intercellular adhesion molecule 1 (ICAM-1). The correlation between 4-GalT-I and ICAM-1 during embryo implantation. 尾 _ 1 ~ (4-galactosyltransferase) I (尾 _ 1N _ 4-GalT-I) distributes in Golgi apparatus and cell membrane, and is located on 尾 _ 1 on Golgi body. 4-GalT-I can perform the glycosylation modification function of protein. As a cell adhesion molecule, 尾 _ 1N _ 4-GalT-I, located on the cell membrane, is involved in cell adhesion, sperm-egg recognition, axon growth, tumor cell migration and so on. Intercellular adhesion molecule-1 (ICAM-1) is an important adhesion molecule. It is also a transmembrane glycoprotein, which plays an important role in the development of early blastocyst, the establishment of endometrial receptivity and the implantation of blastocyst. Methods: human endometrial cancer cells (RL95-2) were used to simulate preimplantation endometrium, and human chorionic carcinoma cells (JARs) were used to simulate blastocyst to form an in vitro implantation model.
