【摘要】: 目的:觀察intermedin (IMD)對大鼠腎臟缺血再灌注損傷(IRI)中氧化應(yīng)激作用的影響。 方法:健康雄性Wistar大鼠30只隨機(jī)分為假手術(shù)組、IRI組、轉(zhuǎn)空質(zhì)粒組、轉(zhuǎn)腎上腺髓質(zhì)素(ADM)質(zhì)粒組、轉(zhuǎn)IMD質(zhì)粒組。動物右腎切除后,用超聲微泡技術(shù)將質(zhì)粒轉(zhuǎn)染入腎臟,1周后制作腎臟IRI模型。半定量分析腎臟病理損傷,比色法檢測腎組織超氧化物岐化酶(SOD)、髓過氧化物酶(MPO)及caspase-3活性、脂質(zhì)過氧化物丙二醛(MDA)含量,免疫組織化學(xué)方法檢測細(xì)胞間粘附分子-1 (ICAM-1)、P-選擇素及內(nèi)皮素1(ET-1)表達(dá),原位末端標(biāo)記法(TUNEL)檢測腎組織細(xì)胞凋亡。 結(jié)果:1、手術(shù)組大鼠發(fā)生急性腎小管壞死,以IRI組病理損傷最重,轉(zhuǎn)染IMD后病理損傷明顯減輕。2、IRI組SOD活性明顯降低,MDA含量明顯增加;MPO活性明顯增高,ICAM-1、P-選擇素表達(dá)明顯增加;ET-1表達(dá)明顯增加;caspase-3活性明顯增高,大量腎小管上皮細(xì)胞凋亡。轉(zhuǎn)染IMD后SOD活性明顯增高,MDA的生成明顯減少;MPO活性明顯降低,ICAM-1、P-選擇素表達(dá)明顯減少;ET-1表達(dá)明顯減少;caspase-3活性明顯降低,腎小管上皮細(xì)胞凋亡減輕,與轉(zhuǎn)ADM組之間沒有明顯差別。轉(zhuǎn)空質(zhì)粒組和IRI組以上指標(biāo)沒有明顯差別。 結(jié)論:IMD在腎臟局部高表達(dá)對腎臟IRI有保護(hù)作用,其機(jī)制至少部分與抑制氧自由基生成、炎細(xì)胞浸潤及炎癥因子ICAM-1、P-選擇素生成,ET-1生成,細(xì)胞凋亡有關(guān),從而減輕腎組織局部氧化應(yīng)激反應(yīng)產(chǎn)生的活性氧族。
[Abstract]:Objective : To observe the effect of intermedin ( IMD ) on oxidative stress in rats with renal ischemia - reperfusion injury ( IRI ) . Methods : 30 healthy male Wistar rats were randomly divided into sham operation group , IRI group , empty plasmid group , adrenomedullin ( ADM ) plasmid group and transferred IMD plasmid group . After the animal right kidney was removed , the plasmid was transfected into the kidney for 1 week . The expression of superoxide dismutase ( SOD ) , myeloperoxidase ( MPO ) , caspase - 3 activity , lipid peroxide malondialdehyde ( MDA ) content and immunohistochemical method were used to detect the expression of intercellular adhesion molecule - 1 ( ICAM - 1 ) , P - selectin and endothelin - 1 ( ET - 1 ) . TUNEL was used to detect the apoptosis of renal tissue cells . Results : 1 . The acute tubular necrosis of renal tubular epithelial cells occurred in the operation group . The expression of SOD in IRI group was significantly decreased , the content of MDA was significantly increased , the expression of ICAM - 1 and P - selectin was significantly increased , the expression of ICAM - 1 and P - selectin was significantly decreased , the expression of ICAM - 1 and P - selectin was significantly decreased , the expression of ICAM - 1 and P - selectin was significantly decreased , the expression of ICAM - 1 and P - selectin was significantly decreased , the expression of ICAM - 1 and P - selectin was significantly decreased , and no significant difference was found between the expression of caspase - 3 and apoptosis of renal tubular epithelial cells . Conclusion : The local high expression of IMD has protective effect on renal IRI , and its mechanism is at least partially related to the inhibition of oxygen free radical formation , inflammatory cell infiltration and inflammatory factor ICAM - 1 , P - selectin formation , ET - 1 formation and apoptosis , thus reducing reactive oxygen species produced by local oxidative stress reaction in renal tissue .
