人胚胎來(lái)源多潛能干細(xì)胞向脂肪和神經(jīng)細(xì)胞分化的研究

【摘要】: 人胚胎肝組織來(lái)源的多潛能干細(xì)胞是從胎兒肝組織中分離的一類(lèi)干細(xì)胞,與較之前報(bào)道的成體干細(xì)胞(如骨髓間充質(zhì)干細(xì)胞)有很多相似的特性,具有多分化潛能并能夠形成新的組織和器官,具有重要的醫(yī)學(xué)治療作用。 本研究對(duì)人胚胎肝組織來(lái)源的多潛能干細(xì)胞進(jìn)行分離和擴(kuò)增,對(duì)其生物學(xué)特性進(jìn)行了研究,探討了人胚胎來(lái)源多潛能干細(xì)胞向脂肪細(xì)胞和神經(jīng)細(xì)胞方向誘導(dǎo)分化的潛能。采用膠原酶消化法分離人胚胎肝組織來(lái)源多潛能干細(xì)胞,經(jīng)過(guò)體外培養(yǎng)和擴(kuò)增,對(duì)人胚胎肝組織來(lái)源多潛能干細(xì)胞的生長(zhǎng)經(jīng)時(shí)變化和表面標(biāo)志CD105、CD166、CD73和HLA-DR進(jìn)行檢測(cè)。應(yīng)用成脂誘導(dǎo)培養(yǎng)液(地塞米松、3-異丁基-1-甲基黃嘌呤、吲哚美辛、胰島素)誘導(dǎo)其向脂肪細(xì)胞分化,使用化學(xué)誘導(dǎo)法誘導(dǎo)其向神經(jīng)細(xì)胞方向分化,然后進(jìn)行鑒定。結(jié)果表明,人胚胎肝組織來(lái)源多潛能干細(xì)胞的生物學(xué)特性與骨髓間充質(zhì)干細(xì)胞類(lèi)似,免疫熒光化學(xué)染色及RT-PCR結(jié)果說(shuō)明人胚胎肝組織來(lái)源多潛能干細(xì)胞經(jīng)誘導(dǎo)可分化成為脂肪細(xì)胞及神經(jīng)細(xì)胞。本研究結(jié)果提示,人胚胎來(lái)源多潛能干細(xì)胞具有分化成脂細(xì)胞和神經(jīng)細(xì)胞的潛能,有望應(yīng)用于組織工程及細(xì)胞治療的種子細(xì)胞。
[Abstract]:Pluripotent human embryonic liver tissue derived stem cells are isolated from liver tissue of the fetus, and compared with the previously reported adult stem cells (such as bone marrow mesenchymal stem cells) have many similar characteristics, with multiple differentiation potential and can form new tissues and organs, with medical treatment an important role.
The study of pluripotent human embryonic liver tissue derived stem cells were isolated and amplified, its biological characteristics were studied, discussed the human embryonic pluripotent stem cells differentiate into fat cells and nerve cells. The direction of the potential separation of human embryonic pluripotent stem cells by collagenase digestion, and after in vitro culture of human embryonic liver tissue CD166 pluripotent stem cell growth by the change and the surface markers CD105, CD73 and HLA-DR were detected. Application of adipogenic induction medium (dexamethasone, 3- -1- isobutyl methylxanthine, indomethacin, insulin) to induce adipocyte differentiation, induction method to induce nerve cell differentiation using chemical, and then were identified. The results showed that human embryonic pluripotent stem cells and biological characteristics of bone marrow mesenchymal stem cells is similar to that of free Immunofluorescence staining and RT-PCR results indicated that human embryonic pluripotent stem cells can be induced into fat cells and nerve cells. The results of this study suggest that human embryonic pluripotent stem cells can differentiate into fat cells and nerve cell potential, is expected to be used as seed cells for tissue engineering and cell therapy.
