四味芳香開竅藥抗急性腦缺血的實(shí)驗(yàn)研究及藥性初探

【摘要】:目的:平行觀察四味芳香開竅藥(麝香、冰片、蘇合香、安息香)抗急性腦缺血的作用,并探討其部分作用機(jī)制,同時(shí)試探急性腦缺血對(duì)模型動(dòng)物心臟的影響,以期闡明該類藥開竅醒神功效和“心—辛—浮”藥性系統(tǒng)的科學(xué)內(nèi)涵,為初步建立類“熱閉證”動(dòng)物模型提供一定實(shí)驗(yàn)依據(jù)。 方法:①采用小鼠斷頭張口實(shí)驗(yàn)、飽和氯化鎂致小鼠急性腦缺血實(shí)驗(yàn)及線栓法阻斷大鼠大腦中動(dòng)脈6h致大鼠局灶性腦缺血模型實(shí)驗(yàn),通過觀察小鼠存活時(shí)間、模型大鼠神經(jīng)功能、腦梗死面積及腦組織形態(tài)學(xué)的變化,評(píng)價(jià)芳香開竅抗急性腦缺血的作用。②測定局灶性腦缺血模型大鼠腦內(nèi)超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量、Na+-K+-ATPase,Ca++-Mg++-ATPase的活力及血清一氧化氮(NO)含量,探討芳香開竅藥抗腦缺血的部分作用機(jī)制。③觀察線栓法致大腦局灶性腦缺血模型大鼠心電圖,心肌組織形態(tài)學(xué)和心SOD活性、MDA含量及Na+-K+-ATPase、Ca++-Mg++-ATPase活力的改變,試探腦缺血與心臟功能的相關(guān)性。④測定模型大鼠體溫、心率與血壓的變化,為復(fù)制類“熱閉證”動(dòng)物模型提供一定實(shí)驗(yàn)依據(jù)。 結(jié)果:①以麝香、冰片為代表的芳香開竅藥能增加斷頭后小鼠的張口次數(shù)與時(shí)間,能延長注射飽和氯化鎂溶液后小鼠的張口喘氣時(shí)間(P0.05)。②線栓法阻斷大鼠大腦中動(dòng)脈后,模型組大鼠蘇醒時(shí)間延長(P0.05),眼口緊閉,病灶對(duì)側(cè)前爪內(nèi)收不能完全伸展,出現(xiàn)神經(jīng)功能損傷(P0.05),并且體溫明顯升高(P0.05)。③與模型組比較,以麝香、冰片為代表的芳香開竅藥能減輕線栓法致大腦局灶性腦缺血大鼠神經(jīng)功能的損傷(P0.05),縮小大鼠局灶性腦缺血梗死面積并減輕其病理改變(P0.05)。④以麝香、冰片為代表的芳香開竅藥能降低線栓法致大腦局灶性腦缺血大鼠腦內(nèi)MDA含量(P0.05),提高腦內(nèi)SOD活性(P0.05);并且還能增加腦缺血模型大鼠腦內(nèi)Na+-K+-ATPase, Ca++-Mg++-ATPase的活力(P0.05),但對(duì)血清NO含量無影響(P0.05)。⑤與假手術(shù)組比較,線栓法致大腦局灶性腦缺血6h大鼠心電圖、心肌組織形態(tài)學(xué)未發(fā)生病理性改變,心肌SOD活性、MDA含量及Na+-K+-ATPase,Ca++-Mg++-ATPase的活力也無明顯變化(P0.05)。 結(jié)論:①以麝香、冰片為代表的芳香開竅藥具有抗急性腦缺血的作用,其機(jī)制可能與降低腦內(nèi)MDA含量、提高腦內(nèi)SOD活性及增加腦缺血大鼠腦內(nèi)Na+-K+-ATPase,Ca++-Mg++-ATPase的活力有關(guān)。②初步建立了類“熱閉證”大鼠模型,四味芳香開竅藥對(duì)模型動(dòng)物體溫、心率及血壓影響的差異反映了該類藥在“四性”上的不同。③芳香開竅藥抗急性腦缺血作用是其開竅醒神功效的藥效表征,也是其“辛—心—浮”藥性系統(tǒng)的藥效譜。④線栓法致大腦局灶性腦缺血6h的大鼠,未能發(fā)現(xiàn)心臟病理性的改變。
[Abstract]:Objective: to observe the effects of four kinds of aromatic orifices (musk, borneol, heparin and benzoin) on acute cerebral ischemia in parallel, and to explore the effects of acute cerebral ischemia on the heart of model animals. The purpose of this paper is to clarify the scientific connotation of this kind of medicine and to provide some experimental basis for the establishment of the animal model of "heat closing syndrome". Methods the mouse head opening test, the acute cerebral ischemia induced by saturated magnesium chloride and the focal cerebral ischemia model of rats by blocking the middle cerebral artery for 6 hours were used. The changes of survival time, neural function, cerebral infarction area and brain histomorphology of the model rats were observed. To evaluate the effect of aromatics on acute cerebral ischemia. 2 to measure the activity of superoxide dismutase (SOD) and the content of malondialdehyde (MDA) in the brain of rats with focal cerebral ischemia. The activity of Ca -Mg ATPase and the content of nitric oxide (no) in serum of Na K ATPase. Objective: to investigate the mechanism of anti-cerebral ischemia by aromatic resuscitation drugs. 3 observe the electrocardiogram (ECG), myocardial morphology and cardiac SOD activity in rats with focal cerebral ischemia induced by thread embolism. The changes of MDA content and Na-K-ATPase activity of Ca-mg-ATPase were studied. The correlation between cerebral ischemia and cardiac function was explored. 4. 4. The temperature of the model rats was measured. The changes of heart rate and blood pressure provide some experimental basis for duplicating the animal model of heat closure syndrome. Results the aromatic orifices represented by musk and borneol could increase the opening times and time of severed mice. After blocking the middle cerebral artery (MCA) of rats by P0.052.Then prolonging the gasping time of mice injected with saturated magnesium chloride solution, the recovery time of rats in the model group was prolonged (P0.05) and the eyes were closed. The adducts of the contralateral anterior claws could not be completely extended, the nerve function was injured and the body temperature was significantly increased compared with the model group. Borneol as the representative aromatic resuscitation medicine can reduce the damage of neural function in rats with focal cerebral ischemia induced by thread embolization (P0.05). The infarct area of focal cerebral ischemia in rats was reduced and its pathological changes were alleviated (P0.05.4) with musk. Borneol as the representative aromatic resuscitation medicine can reduce the content of MDA and increase the activity of SOD in brain of rats with focal cerebral ischemia. It also increased the activity of Na-K-ATPase and Ca-mg-ATPase in the brain of cerebral ischemia model rats (P0.05). Compared with the sham operation group, the electrocardiogram (ECG) of the rats with focal cerebral ischemia induced by thread embolization did not show pathological changes in myocardial histomorphology. The content of SOD and the activity of Ca -Mg -ATPase in Na-K-AT Paseate also had no significant change (P 0. 05). Conclusion the aromatic orifices represented by musk and borneol have the effect of anti-acute cerebral ischemia, and the mechanism may be related to the decrease of MDA content in the brain. The activity of SOD in brain and Na -K ATPase in brain of ischemic rats were increased. The activity of Ca-mg-ATPase was related to the establishment of the rat model of "heat blocking syndrome", and the body temperature of the model animal was treated by four kinds of aromatic orifices. The difference of heart rate and blood pressure reflects that the anti-acute cerebral ischemia effect of three kinds of aromatic resuscitation drugs on "four properties" is a sign of the effect of resuscitation. It was also found that the changes of heart disease were not found in the rats with focal cerebral ischemia induced by the pharmacodynamic spectrum of the "Xin-Xin-Fu" drug system for 6 h.
