腦紅蛋白在感染性休克早期大鼠腦組織中的動(dòng)態(tài)變化

【摘要】:目的探討感染性休克早期大鼠腦組織中腦紅蛋白(neuroglobin,NGB)含量的動(dòng)態(tài)變化在感染性休克中可能發(fā)揮的作用。方法健康雄性SD大鼠60只,12~15周齡,體重300~350g,隨機(jī)分為兩組,每組30只。M組經(jīng)尾靜脈注射脂多糖(LPS)10mg/kg制備大鼠感染性休克模型,當(dāng)SBP下降幅度達(dá)到基礎(chǔ)值的40%時(shí)認(rèn)為感染性休克模型復(fù)制成功;C組給予等量生理鹽水。分別在給LPS前(T_0)、給LPS后1h(T_1)、2h(T_2)、4h(T_3)、6h(T_4)采用ELISA法檢測(cè)血清中TNF-α、IL-6濃度和NGB含量,Western blot法檢測(cè)腦組織中NGB、半胱天冬酶-3(caspase-3)的含量。結(jié)果與C組比較,T1~T4時(shí)M組血清中的TNF-α、IL-6濃度明顯增高(P0.05),腦組織中caspase-3含量明顯增加(P0.05);T_3、T_4時(shí)血清和腦組織中NGB含量明顯增高(P0.05)。結(jié)論感染性休克后大鼠腦組織發(fā)生了炎癥損傷、細(xì)胞凋亡病理學(xué)改變,腦紅蛋白在感染性休克所致的損傷腦組織中含量增加,推測(cè)其可能發(fā)揮了內(nèi)源性神經(jīng)保護(hù)功能。
[Abstract]:Objective to investigate the role of the dynamic changes of brain erythroprotein neuroglobinus NGB (NGB) in the early stage of septic shock in rats. Methods 60 healthy male Sprague-Dawley rats were randomly divided into two groups. The septic shock model of rats in group M was induced by injecting lipopolysaccharide (LPS) 10 mg / kg into tail vein of 30 rats in each group. When the decrease of SBP reached the base value, it was considered that the septic shock model was successfully replicated and group C was given the same amount of normal saline. Before LPS was given, T0 was given, and 1 hour after LPS was given, respectively, 2hstr T2T 2hStaph, 4hStaph T3hT 3n, 6hStam T4). ELISA method was used to detect TNF- 偽 -IL-6 and NGB content. Western blot method was used to detect the concentration of TNF- 偽, IL-6 and NGB. Results compared with group C, the concentration of TNF- 偽 and IL-6 in serum of group M was significantly higher than that in group C (P 0.05), and the content of caspase-3 in brain tissue was significantly higher than that in group C. Conclusion the content of NGB in serum and brain tissue is significantly higher than that in group C. Conclusion the content of NGB in serum and brain tissue is significantly higher than that in group C. Conclusion the level of NGB in serum and brain tissue of group M is significantly higher than that of group C. Conclusion the level of NGB in serum and brain tissue of group M is significantly higher than that of group C. Inflammatory injury occurred in the brain of the rats after sexual shock. The pathological changes of apoptosis and the increase of the content of brain erythroprotein in the injured brain tissue caused by septic shock suggest that it may play an endogenous neuroprotective function.
