YAP,β-catenin在乳腺浸潤性導管癌中表達及其臨床意義

【摘要】:目的:分別檢測乳腺浸潤性導管癌(非特殊型)、乳腺腺瘤以及正常乳腺組織中YAP和β-catenin的表達情況,以此分析YAP、β-catenin二者與乳腺浸潤性導管癌臨床病理因素之間的關系、表達的相關性、與乳腺浸潤性導管癌預后的關系,進一步探討YAP、β-catenin兩個蛋白與乳腺浸潤性導管癌的發(fā)生、發(fā)展及預后的關系。方法:采用免疫組化SP法檢測YAP和β-catenin在32例正常乳腺組織、40例乳腺腺瘤組織及97例乳腺浸潤性導管癌組織中的表達情況。結果:(1)YAP在正常乳腺組織中的表達明顯高于乳腺腺瘤組織及乳腺浸潤性導管癌組織,其在正常乳腺組織、乳腺腺瘤組織及乳腺浸潤性導管癌組織中的陽性表達率依次減低,差異有統(tǒng)計學意義(P0.05)。在乳腺浸潤性導管癌組織中,β-catenin的表達情況明顯高于在正常乳腺組織的表達,也高于在乳腺腺瘤組織的表達。β-catenin在正常乳腺組織、乳腺腺瘤組織及乳腺浸潤性導管癌組織中的陽性表達率依次上調,差異有統(tǒng)計學意義(P0.05)。(2)YAP表達與乳腺浸潤性導管癌患者年齡、組織學分級、腫瘤大小、淋巴結轉移情況、是否絕經等均無相關性(P0.05);與是否為三陰性乳腺癌相關(P0.05)。β-catenin表達與患者年齡、腫瘤大小、是否絕經等均無相關性(P0.05);與組織學分級、淋巴結轉移情況、是否為三陰性乳腺癌相關(P0.05)。(3)YAP的陽性表達與乳腺浸潤性導管癌患者的總生存率不相關(P0.05);與無復發(fā)生存率相關(P0.05)。β-catenin陽性表達與乳腺浸潤性導管癌患者的總生存率、無復發(fā)生存率均不相關(P0.05)。YAP(-)/β-catenin(+)共表達與乳腺浸潤性導管癌患者的總生存率、無復發(fā)生存率均相關(P0.05)。(4)在乳腺浸潤性導管癌、乳腺腺瘤組織中YAP和β-catenin的表達均呈負相關(rs為負值,P0.01);YAP和β-catenin二者在正常乳腺中的表達無相關性(P0.05)。結論:(1)β-catenin在乳腺浸潤性導管癌組織中呈高表達,并與淋巴結轉移情況相關,提示βcatenin對乳腺浸潤性導管癌的發(fā)生和轉移有促進作用。(2)β-catenin在乳腺浸潤性導管癌組織中的表達情況與組織分級有關,提示β-catenin可作為乳腺浸潤性導管癌組織分級判斷的參考指標。(3)在乳腺浸潤性導管癌組織中YAP和β-catenin的表達呈負相關,且YAP(-)/β-catenin(+)共表達與乳腺浸潤性導管癌患者的總生存率、無復發(fā)生存率均相關,提示二者聯(lián)合檢測有助于乳腺浸潤性導管癌的臨床診斷、療效評估及預后判斷。
[Abstract]:Objective: to detect the expression of YAP and 尾 -catenin in invasive ductal carcinoma of breast (non-special type, adenoma and normal mammary gland), and to analyze the relationship between YAPand 尾 -catenin and clinicopathological factors of invasive ductal carcinoma of breast.The relationship between YAPand 尾 -catenin protein and the occurrence, development and prognosis of invasive ductal carcinoma of breast were discussed.Methods: immunohistochemical SP method was used to detect the expression of YAP and 尾 -catenin in 32 cases of normal breast tissues, 40 cases of breast adenoma and 97 cases of breast invasive ductal carcinoma.Results the expression of yap in normal breast tissue was significantly higher than that in adenoma tissue and invasive ductal carcinoma tissue, and the positive expression rate in normal breast tissue, breast adenoma tissue and invasive ductal carcinoma tissue was decreased in turn.The difference was statistically significant (P 0.05).The expression of 尾 -catenin in breast invasive ductal carcinoma was significantly higher than that in normal breast and adenoma.The positive expression rate in breast adenoma and breast invasive ductal carcinoma was up-regulated in turn. The difference was statistically significant between the expression of YAP and age, histological grade, tumor size, lymph node metastasis in patients with invasive ductal carcinoma of breast.The expression of 尾 -catenin was not correlated with age, tumor size, menopause, histological grade and lymph node metastasis.There was no correlation between the positive expression of P0.05A and the overall survival rate of breast invasive ductal carcinoma patients, and the correlation between the positive expression of 尾 -catenin and the overall survival rate of breast infiltrating ductal carcinoma patients was not correlated with the recurrence free survival rate (P 0.05). The positive expression of 尾 -catenin was associated with the overall survival rate of invasive ductal carcinoma of the breast.The recurrence free survival rate was not correlated with the overall survival rate of breast invasive ductal carcinoma patients, and the recurrence free survival rate was correlated with the P0.05P / 尾 -catenin (尾 -catenin (尾 -catenin) co-expression) in invasive ductal carcinoma of the breast.The expression of YAP and 尾 -catenin in breast adenoma was negatively correlated with the expression of 尾 -catenin. There was no correlation between the expression of YAP and 尾 -catenin in normal mammary gland.Conclusion 尾 -catenin is highly expressed in breast invasive ductal carcinoma, and is related to lymph node metastasis.It is suggested that 尾 catenin can promote the occurrence and metastasis of invasive ductal carcinoma of breast. The expression of 尾 -catenin in breast invasive ductal carcinoma is related to the histological grade.The results suggest that 尾 -catenin can be used as a reference index for grading of invasive ductal carcinoma of breast. The expression of YAP and 尾 -catenin in invasive ductal carcinoma of breast is negatively correlated with the expression of 尾 -catenin (YAP(-)/ 尾 -catenin), and the overall survival rate of patients with invasive ductal carcinoma of breast is associated with the co-expression of YAP(-)/ 尾 -catenin (YAP(-)/ 尾 -catenin (尾 -catenin)).The recurrence free survival rate was correlated, suggesting that the combined detection was helpful for the clinical diagnosis, evaluation of curative effect and prognosis of invasive ductal carcinoma of breast.
