溶血磷脂酸影響胚胎神經(jīng)干細(xì)胞增殖和分化的初步觀察

  神經(jīng)干細(xì)胞(neural stem cells,NSCs)廣泛存在于胚胎腦組織中,也存在于成年哺乳動物的神經(jīng)系統(tǒng)。NSCs具有自我更新能力,能夠定向分化為神經(jīng)元、星形膠質(zhì)細(xì)胞和少突膠質(zhì)細(xì)胞。NSCs的這一特點使其有望成為體外移植治療神經(jīng)系統(tǒng)損傷和退行性疾病的重要材料。 溶血磷脂酸(lysophospatidic acid,LPA)是一種簡單的磷脂類物質(zhì),是血清的正常成份,而且正常情況下也存在于腦組織內(nèi)。LPA的受體,LPAl-3,在多種細(xì)胞的細(xì)胞膜上有不同表達。LPA與G蛋白耦聯(lián)的受體結(jié)合后,激活下游信號傳導(dǎo)通路,從而調(diào)節(jié)體內(nèi)各種細(xì)胞,包括神經(jīng)元、神經(jīng)膠質(zhì)細(xì)胞等的功能活動。LPA在細(xì)胞水平的作用很廣泛,主要是影響細(xì)胞形態(tài),促進細(xì)胞增殖和存活,誘導(dǎo)細(xì)胞凋亡,改變離子電流和鈣胞內(nèi)釋放等。Contos等曾報道,1.0 μmo/L LPA可以促進小鼠胚胎NSCs的增殖。但也有文獻報道LPA的作用具有兩面性,即不同濃度的LPA對同一細(xì)胞的功能影響不同;而且,LPA對來自大鼠胚胎的NSCs的增殖和分化方面的影響如何,并未見有關(guān)報道。因此,本實驗第一部分探討了LPA對體...
