小鼠軟腦膜黑色素瘤原位模型的建立及其在評價(jià)腦腫瘤疫苗中的應(yīng)用

軟腦膜黑色素瘤(LMM),從顱外黑色素瘤的轉(zhuǎn)移灶或從本有的黑色素細(xì)胞開始發(fā)展,在預(yù)后差的黑色素瘤患者中,有22%的發(fā)病率。在本實(shí)驗(yàn)研究中,我們建立了一個(gè)小鼠大腦LMM模型,在C57BL/6小鼠的頭骨上打孔,使用一個(gè)自我改良的微量注射器將B16細(xì)胞直接注入到蛛網(wǎng)膜下腔。35%至40%的小鼠大腦或大腦和小腦延髓池浸潤黑色素瘤,.10%的小鼠大腦或小腦浸潤,15%的小鼠枕大池和脊髓浸潤。黑色素瘤在大腦半球的表面形成了一個(gè)約1.5×1.3×1.0厘米大小的黑色膠狀團(tuán)塊,有多處隆起并聚集的小瘤塊。在腦基底部的柔腦膜、大腦和脊髓無可見黑色素瘤。組織病理學(xué)檢測可見黑色素瘤組織充滿B16細(xì)胞且血管生長迅速,腦組織無黑色素瘤細(xì)胞,黑色素細(xì)胞廣泛地滲透在軟腦膜層。在臨床上,在小鼠身上發(fā)現(xiàn)的腫瘤位置和可檢測的癥狀的相關(guān)性,在患者LMM是同樣適用的。為了測試的小鼠模型的可行性,我們也嘗試了用腫瘤細(xì)胞B16裂解物(TCL)免疫LMM模型小鼠去誘導(dǎo)抗LMM免疫。
