NO對(duì)大鼠DRG神經(jīng)元胞體膜的作用

目的:本實(shí)驗(yàn)以SD大鼠背根神經(jīng)節(jié)(dorsal root ganglion, DRG)為模型,觀察一氧化氮(nitric oxide, NO)對(duì)DRG神經(jīng)元的作用及有關(guān)離子機(jī)制,并就NO在痛覺(jué)信息傳遞過(guò)程中的作用進(jìn)行了探討,為臨床痛覺(jué)的治療提供一些新的思路。方法: 1、在分離的大鼠新鮮DRG標(biāo)本上,應(yīng)用細(xì)胞內(nèi)記錄技術(shù),給予灌流NO供體硝普鈉(sodium nitroprusside, SNP),觀察NO對(duì)DRG神經(jīng)元的作用。2、灌流SNP記錄膜電位變化的同時(shí),經(jīng)記錄膜電位的同一微電極注入超極化方波(波寬400 ms,強(qiáng)度1.0 nA,頻率0.25 Hz),根據(jù)胞膜緊張電位的變化而推算出膜電導(dǎo)值的改變。3、利用通道阻斷劑和無(wú)Na+平衡鹽液(balance salt solution, BSS)的方法,觀察NO作用的離子機(jī)制。結(jié)果:1、大部分DRG神經(jīng)元對(duì)SNP敏感(79/102, 77.45%),施加SNP(10~100 mmol/L)后可引起濃度依賴(lài)性的超極化反應(yīng),EC50值為30.3 mmol/L。剩余神經(jīng)元沒(méi)有反應(yīng)。100 mmol/L SNP可使神經(jīng)元膜電導(dǎo)由21.06±1.94...
