JAK2/STAT通路在人腎小球系膜細(xì)胞衰老過程中的作用研究

目的腎臟是機(jī)體衰老的主要器官,腎小球?yàn)V過率(glomerular filtration rate,GFR)隨增齡而降低,進(jìn)入45歲以后GFR下降速度加快。基于隨著經(jīng)濟(jì)發(fā)展老齡人口逐漸增加及老年人是慢性腎臟病高危人群這兩方面因素,在我國(guó)逐漸邁入老齡化社會(huì)的今天,研究腎臟衰老機(jī)制、探討延緩腎臟衰老的方法,顯得尤為重要。腎臟衰老的機(jī)制及防治是目前國(guó)內(nèi)外學(xué)者關(guān)注的重要問題之一。目前關(guān)于腎臟衰老的機(jī)制尚不十分明確。多數(shù)學(xué)者認(rèn)為腎臟衰老主要是由于腎臟固有細(xì)胞功能減退所致。腎小球系膜細(xì)胞是腎臟的主要固有細(xì)胞,其表型和功能改變?cè)谀I臟衰老進(jìn)程中發(fā)揮著重要的作用。目前的研究證實(shí),高糖、血管緊張素Ⅱ(angiotensin, AngⅡ)、叔丁基過氧化氫(tert-butly hydroperoxide, tBHP)可促進(jìn)細(xì)胞衰老。這些研究多見于臍靜脈血管內(nèi)皮細(xì)胞、肺成纖維細(xì)胞、腹膜間皮細(xì)胞、血管平滑肌細(xì)胞等,尚沒有對(duì)人腎小球系膜細(xì)胞(Human Glomerular Mesangial Cell, HGMC)衰老的研究報(bào)道。JAK2/STAT通路是一條重要的信號(hào)轉(zhuǎn)導(dǎo)途徑,在心肌細(xì)胞、巨噬細(xì)胞、T細(xì)胞等多種細(xì)...
