BMP-2、VEGF165雙基因共表達質(zhì)粒在小鼠骨髓基質(zhì)干細胞的表達

【摘要】: 目的:觀察轉(zhuǎn)染BMP-2和VEGF165雙基因共表達質(zhì)粒在小鼠骨髓基質(zhì)干細胞的表達。方法:脂質(zhì)體介導下將雙基因真核表達質(zhì)粒pIRES-BMP2-VEGF165導入小鼠骨髓基質(zhì)干細胞,用RT-PCR和免疫組織化學方法觀察BMP-2和VEGF165雙基因在小鼠骨髓基質(zhì)干細胞內(nèi)的表達。結(jié)果:①傳代后的mBMSC_S增殖迅速,5-7 d細胞達90%融合。傳代后不再以集落方式生長,呈均勻分布生長,細胞形態(tài)均勻,與成纖維細胞形態(tài)相似,大部分呈長梭形,排列規(guī)則。②轉(zhuǎn)染BMP-2、VEGF165的小鼠骨髓基質(zhì)干細胞有明顯的BMP-2和VEGF165 mRNA及其蛋白表達。結(jié)論:經(jīng)轉(zhuǎn)染BMP-2和VEGF165雙基因質(zhì)粒的小鼠骨髓基質(zhì)干細胞能同時表達以上兩種基因。
[Abstract]:Aim: to observe the expression of BMP-2 and VEGF165 co-expression plasmid in mouse bone marrow stem cells (BMSCs). Methods: the double gene eukaryotic expression plasmid pIRES-BMP2-VEGF165 was introduced into mouse bone marrow stroma stem cells mediated by liposomes. The expression of BMP-2 and VEGF165 genes in mouse bone marrow stroma stem cells was observed by RT-PCR and immunohistochemistry. Results: 1 the mBMSC_S proliferated rapidly after passage, and 90% of the cells were fused on the 7th day. After passage, it no longer grows in the form of colony, showing uniform distribution and growth, and the morphology of cells is similar to that of fibroblasts, most of which are long fusiform and regular arrangement. 2 the expression of BMP-2, VEGF165 mRNA and their proteins were obvious in mouse bone marrow stroma stem cells transfected with BMP-2,VEGF165. Conclusion: mouse bone marrow stroma stem cells transfected with BMP-2 and VEGF165 double gene plasmid can express the above two genes at the same time.
