樹(shù)突狀細(xì)胞共刺激分子表達(dá)與免疫抑制劑的干預(yù)

【摘要】:背景:以往關(guān)于器官移植免疫抑制和抗排斥反應(yīng)的研究,多關(guān)注T淋巴細(xì)胞介導(dǎo)的免疫反應(yīng)及免疫抑制劑對(duì)T淋巴細(xì)胞的作用,樹(shù)突狀細(xì)胞作用尚不清楚,且對(duì)于免疫抑制劑對(duì)樹(shù)突狀細(xì)胞影響的表現(xiàn)及機(jī)制亦不盡相同。目的:比較不同免疫抑制劑對(duì)樹(shù)突狀細(xì)胞共刺激分子表達(dá)及功能的影響,探討其免疫抑制作用機(jī)制。方法:在誘導(dǎo)C57BL/6小鼠骨髓細(xì)胞定向分化為樹(shù)突狀細(xì)胞時(shí)分別加入20μg/L雷帕霉素、0.04 mg/L霉酚酸酯、10μg/L他克莫司和1 mg/L環(huán)孢霉素A。結(jié)果與結(jié)論:流式細(xì)胞儀檢測(cè)結(jié)果顯示,各組CD40表達(dá)為:雷帕霉素他克莫司環(huán)孢霉素A霉酚酸酯(P0.01);CD86表達(dá)分別為:雷帕霉素他克莫司環(huán)孢霉素A=霉酚酸酯(P0.01);CD80、CD11c及主要組織相容性復(fù)合體Ⅱ的表達(dá)各組差異無(wú)顯著性意義(P0.05)。單向混合淋巴細(xì)胞反應(yīng)結(jié)果顯示,各組樹(shù)突狀細(xì)胞共刺激T細(xì)胞增殖能力表現(xiàn)為:雷帕霉素霉酚酸酯他克莫司=環(huán)孢霉素A(P0.05)。結(jié)果證實(shí),雷帕霉素、他克莫司、環(huán)孢霉素A、霉酚酸酯均通過(guò)抑制樹(shù)突狀細(xì)胞共刺激分子CD40及CD86表達(dá)而發(fā)揮免疫抑制作用,以雷帕霉素最為顯著;雖然以上藥物對(duì)樹(shù)突狀細(xì)胞抗原遞呈能力主要組織相容性復(fù)合體Ⅱ的表達(dá)均無(wú)明顯抑制性,但均能抑制樹(shù)突狀細(xì)胞刺激T細(xì)胞增殖的能力,雷帕霉素的抑增殖能力最強(qiáng)。
