【摘要】：背景:人表皮干細(xì)胞和汗腺細(xì)胞的分離培養(yǎng)及鑒定,為探討人表皮干細(xì)胞再生汗腺的可行性打下基礎(chǔ)。目的:探討體外分離培養(yǎng)人表皮干細(xì)胞及汗腺細(xì)胞的有效方法。方法:采集不同年齡段的泌尿外科患者術(shù)后包皮組織,充分清洗消毒后去除皮下組織,將其修剪成0.5cm×0.5cm的皮片,用Ⅳ型膠原純化、富集成人表皮干細(xì)胞,倒置相差顯微鏡觀察細(xì)胞形態(tài):使用免疫熒光染色對(duì)成人表皮干細(xì)胞表型進(jìn)行分析;CCK-8檢測(cè)細(xì)胞的增殖曲線;采用膠原酶消化法從人全層無(wú)損傷皮膚中分離提取汗腺細(xì)胞,并進(jìn)行擴(kuò)增和鑒定。結(jié)果:倒置相差顯微鏡下見分離培養(yǎng)的成人表皮干細(xì)胞呈卵圓形、細(xì)胞之間緊密相連,呈鋪路石狀,免疫熒光染色顯示細(xì)胞表達(dá)CK19、β1整合素。倒置相差顯微鏡下見汗腺細(xì)胞呈扁平多角形,表達(dá)汗腺細(xì)胞標(biāo)志細(xì)胞CK7、18、19及CEA。結(jié)論:本實(shí)驗(yàn)結(jié)果說(shuō)明膠原酶消化法分離培養(yǎng)成人表皮干細(xì)胞及汗腺樣細(xì)胞是可行的。
