減體積肝移植模型大鼠肝臟能量差異蛋白的表達(dá)

【摘要】:背景:目前,蛋白質(zhì)組學(xué)是一項比較成熟的科研技術(shù),已經(jīng)在肝移植基礎(chǔ)研究領(lǐng)域中初步得到應(yīng)用,但在大鼠減體積肝移植相關(guān)研究中的應(yīng)用未曾報道。目的:應(yīng)用蛋白質(zhì)組學(xué)相關(guān)技術(shù)探討大鼠減體積肝移植后與肝臟能量代謝蛋白相關(guān)的差異蛋白。方法:肝移植的供體是Lewis雌性大鼠,受體是Wistar雄性大鼠,供受體體質(zhì)量差約20 g;移植肝質(zhì)量/受體肝質(zhì)量≈50%。采用改良減體積肝移植模型。獲取供體肝組織過程中按照要求進(jìn)行減體積,修整供體肝臟,將門靜脈和肝下下腔靜脈分別套入不同型號的套管以備套入受體的門靜脈和肝下下腔靜脈,膽道用細(xì)小支撐管植入供體的膽管中以備插入到受體的膽管中;最后進(jìn)行受體的肝移植,受體先切除受體的原來肝臟組織,然后將準(zhǔn)備好的供體肝臟植入受體中。造模后1,3和7 d獲取移植肝臟組織,然后與預(yù)先獲取凍存的供、受體肝臟組織應(yīng)用蛋白組學(xué)技術(shù)建立雙向凝膠電泳圖譜,再利用串聯(lián)質(zhì)譜分析及數(shù)據(jù)庫對差異表達(dá)的蛋白質(zhì)點進(jìn)行鑒定。結(jié)果與結(jié)論:采用變化倍數(shù)大于10倍為標(biāo)準(zhǔn)進(jìn)行差異蛋白點的選擇,結(jié)果總共發(fā)現(xiàn)了72個差異點,通過鑒定,最終32個蛋白的功能比較明確,其中有ATP合成酶β亞基、電子轉(zhuǎn)化黃素蛋白β多肽和質(zhì)子轉(zhuǎn)運ATP合成酶3個蛋白參與了細(xì)胞能量代謝的過程,其分布于肝移植后的第1和7天,占6%。
