表皮干細(xì)胞對(duì)角質(zhì)形成細(xì)胞增殖的影響

【摘要】: [目的]表皮干細(xì)胞作為皮膚發(fā)生修復(fù)重建的關(guān)鍵源泉,其生物學(xué)作用尚不清楚。本實(shí)驗(yàn)通過探討表皮干細(xì)胞對(duì)角質(zhì)形成細(xì)胞的黏附、增殖等生物學(xué)行為的影響,進(jìn)一步揭示表皮干細(xì)胞在組織修復(fù)過程中可能的生物學(xué)作用。 [方法]以Ⅳ膠原作為底物,培養(yǎng)表皮細(xì)胞。利用Ⅳ膠原的快速黏附法分離培養(yǎng)表皮干細(xì)胞。然后利用免疫細(xì)胞化學(xué)分別對(duì)角蛋白19和β1整合素進(jìn)行鑒別。將制備的表皮細(xì)胞懸液接種在包被有Ⅳ膠原24孔板上(共6板)。隨機(jī)分為2組:A組為未棄掉表皮干細(xì)胞組,B組為棄掉表皮干細(xì)胞組。制備好表皮細(xì)胞懸液后,調(diào)整細(xì)胞密度為1×10~5/ml,接種于鋪有Ⅳ膠原的24孔板上。溫箱內(nèi)孵育15分鐘后吸掉其中B組的上清液,調(diào)整細(xì)胞密度為1×10~5/ml后接種在24孔板上繼續(xù)作角質(zhì)形成細(xì)胞的培養(yǎng)。分別取培養(yǎng)1天、2天、3天、4天、5天、6天、7天、8天細(xì)胞每天取4孔在顯微鏡下進(jìn)行細(xì)胞計(jì)數(shù),取其平均值,繪制生長(zhǎng)曲線圖。 [結(jié)果]培養(yǎng)出來的表皮干細(xì)胞通過鑒定角蛋白19和β1整合素均有陽性表達(dá);顯微鏡下可見A組(未棄掉表皮干細(xì)胞組)細(xì)胞在第2天即可看到大部分細(xì)胞貼壁,第3天可見細(xì)胞成集落樣生長(zhǎng),且在第6天時(shí)大部分細(xì)胞連成片;而B組(棄掉表皮干細(xì)胞組)細(xì)胞貼壁相對(duì)緩慢,且形成的集落小而少。通過繪制生長(zhǎng)曲線可見A組(未棄掉表皮干細(xì)胞組)細(xì)胞較B組(棄掉表皮干細(xì)胞組)細(xì)胞生長(zhǎng)速度要快,并且在各時(shí)像點(diǎn)細(xì)胞數(shù)均高于對(duì)照組。 [結(jié)論]表皮干細(xì)胞對(duì)角質(zhì)形成細(xì)胞的增殖有正性調(diào)節(jié)作用,表皮干細(xì)胞可能通過分泌某種“表皮干細(xì)胞因子”對(duì)角質(zhì)形成細(xì)胞發(fā)揮作用。
