基于P38MAPK探討隔藥餅灸對慢性支氣管炎模型大鼠肺組織TNF-α的影響及作用機理

【摘要】:目的:觀察隔藥餅灸對慢性支氣管炎大鼠肺組織中TNF-α、P-ATF2的影響,探討其作用機制,為該法防治慢支提供實驗依據(jù)。 方法:將48只SPF級健康SD大鼠隨機分為A(空白對照組)、B(慢支模型組)、C(隔藥餅灸組)、D(隔紙墊灸組)四組,每組12只。采用改良煙熏法復(fù)制慢支大鼠模型。以隔藥餅灸為處理因素,與空白組、模型組、隔紙墊灸組進行對照,取材分兩次進行(即治療前與治療后),測定每組大鼠支氣管肺泡灌洗液中(BALF)中巨噬細(xì)胞含量及肺組織中TNF-α、P-ATF2的含量變化,探討隔藥餅灸對P38MAPK調(diào)控CB大鼠免疫功能的影響。 結(jié)果:(1)大鼠肺組織病理學(xué)變化:隔藥餅灸和隔紙墊灸都有控制炎癥的作用,隔藥餅灸效果更明顯。(2)大鼠BALF中細(xì)胞計數(shù)及AM(肺泡巨噬細(xì)胞)分類計數(shù):隔藥餅灸和隔紙墊灸能降低慢支大鼠BALF細(xì)胞總數(shù)及AM計數(shù)(P0.05);隔藥餅灸效果更好(P0.05)。(3)大鼠肺組織P-ATF2表達(dá):隔藥餅灸和隔紙墊灸均能降低慢支大鼠肺組織P-ATF2含量(P0.05);隔藥餅灸組效果更明顯(P0.05)。(4)大鼠肺組織TNF-α mRNA及TNF-α蛋白表達(dá):隔藥餅灸和隔紙墊灸均能降低慢支大鼠肺組織TNF-α mRNA及TNF-α蛋白表達(dá)(P0.05);隔藥餅灸效果更好(P0.05)。(5)治療前大鼠肺組織P-ATF2表達(dá)分別與TNF-αmRNA、TNF-α蛋白表達(dá)都呈正相關(guān)(r=0.597, r=0.642, P0.01);治療后大鼠肺組織P-ATF2表達(dá)分別與TNF-αmRNA、TNF-α蛋白表達(dá)都呈正相關(guān)(r=0.682, r=0.763, P<0.01)。 結(jié)論:(1)隔藥餅灸可以降低CB大鼠BALF中AM含量,減輕炎癥反應(yīng)。(2)隔藥餅灸能降低大鼠肺組織中TNF-α及P-ATF2含量。且P-ATF2在慢支的炎癥反應(yīng)中起著重要作用,隔藥餅灸通過降低P-ATF2含量而減少其對TNF-α的激活,從而改善慢支炎癥。(3)隔藥餅灸和隔紙墊灸都能改善慢支大鼠的癥狀,但隔藥餅灸效果更好。
[Abstract]:Objective: to observe the effect of herbal cake moxibustion on TNF- 偽 and P-ATF2 in lung tissue of rats with chronic bronchitis, to explore its mechanism, and to provide experimental basis for the prevention and treatment of chronic bronchitis by this method. Methods: Forty-eight healthy SD rats of SPF grade were randomly divided into four groups: group A (control group,), B (slow branch model group), C (separated cake moxibustion group,), D (paper-separated moxibustion group, 12 rats in each group. The rat model of chronic bronchitis was established by modified smoking method. Taking medicine-separated cake moxibustion as treatment factor, it was compared with blank group, model group and paper-separated moxibustion group. The contents of macrophages in bronchoalveolar lavage fluid (BALF) and changes of TNF- 偽 and P-ATF2 in lung tissue were measured in each group. The effect of moxibustion on the immune function of CB rats was studied. Results: (1) pathological changes of lung tissue in rats: both medicine-separated moxibustion and paper-separated moxibustion could control inflammation. (2) the cell count and AM (alveolar macrophage) count in rat BALF: moxibustion and paper-separated moxibustion could reduce the BALF cell count and AM count in slow branch rats (P0.05); The effect of medicine-separated cake moxibustion was better (P0.05). (3) the expression of P-ATF2 in lung tissue of rats was better than that of the control group (P0.05). The content of P-ATF2 in lung tissue of rats with slow bronchitis was decreased by the moxibustion of separated medicine cake and paper-separated moxibustion (P0.05). The expression of TNF- 偽 mRNA and TNF- 偽 protein in lung tissue was more obvious in the moxibustion group (P0.05). (4). The expression of TNF- 偽 mRNA and TNF- 偽 protein in the lung tissue of the chronic bronchitis rats was decreased by the moxibustion with the separated medicine cake and the paper-separated moxibustion (P0.05). The effect of moxibustion with medicine separated cake was better (P0.05). (5) the expression of P-ATF2 in lung tissue was positively correlated with the expression of TNF- 偽 mRNA,TNF- 偽 protein before treatment (r 0.597,0.642, P0.01), and the expression of P-ATF2 in lung tissue was positively correlated with the expression of TNF- 偽 mRNA,TNF- 偽 protein after treatment (r 0.682, r 0.763, P < 0.01). Conclusion: (1) moxibustion with medicine-separated cake can decrease the content of AM in BALF of CB rats and alleviate the inflammatory reaction. (2) moxibustion with separated medicine cake can decrease the content of TNF- 偽 and P-ATF2 in lung tissue of rats. P-ATF2 plays an important role in the inflammatory reaction of the slow bronchitis, and the activation of TNF- 偽 is reduced by decreasing the content of P-ATF2. (3) both the moxibustion of separated medicine cake and the moxibustion with paper partition can improve the symptoms of the rats with chronic bronchitis. But the effect of moxibustion with separated medicine cake is better.
