人牙根尖乳頭細(xì)胞體外無(wú)支架三維培養(yǎng)的生物學(xué)行為研究

【摘要】:目的探討根尖乳頭干細(xì)胞(stem cells from the apical papilla,SCAP)在無(wú)支架三維培養(yǎng)條件下的生物學(xué)行為。方法用顯微鏡和HR染色觀察SCAP球的大體形態(tài),組織學(xué)結(jié)構(gòu);用免疫組化法檢測(cè)SCAP球中骨涎蛋白(bone sialoprotein,BSP)和血管性血友病因子(von Willebrand factor,vWF)的表達(dá)。結(jié)果細(xì)胞球的大小對(duì)球體內(nèi)部細(xì)胞的生長(zhǎng)有影響,SCAP球具有向成骨和向內(nèi)皮分化的潛能。結(jié)論無(wú)支架三維培養(yǎng)是一種可行的SCAP培養(yǎng)方法。
[Abstract]:Objective to investigate the biological behavior of apical papilla stem cells (stem cells from the apical papilla,SCAP) in three-dimensional culture without scaffold. Methods the gross morphology and histological structure of SCAP balls were observed by microscope and HR staining, and the expression of bone sialoprotein,BSP and (von Willebrand factor,vWF in SCAP spheres were detected by immunohistochemistry. Results the size of the spheres had an effect on the growth of the cells inside the spheres, and the SCAP spheres had the potential of osteogenesis and endothelial differentiation. Conclusion three-dimensional culture without scaffold is a feasible method for SCAP culture.
