人體腹部脂肪中解偶聯(lián)蛋白1(UCP1)基因表達(dá)水平的探索研究
[Abstract]:Aim: to maintain the body temperature in cold environment or to cause excess nutrition by food energy, the energy consumption is mainly through the heat production function of brown adipose tissue. Both mechanisms were derived from a specific protein, conjugate protein 1 (uncoupling protein 1 (UCP1). UCP1 was expressed only in mitochondria in brown adipocytes. This study was to investigate the presence of UCP1, in adult abdominal fat (including subcutaneous and visceral) to verify the presence of brown adipose tissue (brown adipose tissue, BAT). In white adipose tissue (white adipose tissue, WAT) in adults. To further investigate the relationship between UCP1 gene expression and lipid index, body mass index, waist circumference and basal metabolism in adult abdominal subcutaneous and visceral adipose tissue. Methods: from August 2008 to January 2009, 25 patients who were newly admitted to the second department of the people's Hospital of Xinjiang Uygur Autonomous region and who needed to undergo open surgery were studied. Subcutaneous and visceral fresh adipose tissue were obtained 3 g and 3 g respectively. RT-PCR technique was used to detect the expression and expression level of UCP1 gene in human abdominal adipose tissue. The relationship between gene expression in brown adipose tissue and energy metabolism and obesity was studied. UCP1 amplified primer sequence was designed to refer to the known Genbank sequence (Gene ID:24860, ref NM_021833.3). To further analyze the expression of UCP1mRNA gene in adult abdominal adipose tissue, including height, body weight, body mass index, waist-hip circumference, systolic blood pressure, diastolic blood pressure, fasting plasma glucose, total cholesterol, triglyceride, high density lipoprotein. Relationship between insulin resistance index and basal metabolism. Results: a total of 25 subjects (13 males) with an average age of (44.68 鹵10.93) years were enrolled in the study. Among the 25 subjects, 21 cases were successfully amplified. The gray value ratio of UCP1/ 尾 -actin in subcutaneous adipose tissue and visceral adipose tissue was (0.79 鹵0.12) and (0.82 鹵0.11), respectively. The expression of UCP1 gene in abdominal adipose tissue was correlated with body mass index (BMI),) height, weight, waist circumference, blood pressure, total cholesterol (TC), triglyceride, (TG), (GLU), high density lipoprotein (HDL), fasting plasma glucose and (FPG). But all did not reach the significant level. Only the insulin resistance index (HOMA-IR) was negatively correlated with the expression of UCP1 gene (r-0.700). Conclusion: the results of this study confirm the presence of brown fat in adult abdominal fat, the expression of brown fat in subcutaneous and visceral white adipose tissue of adult abdomen is not different, and the expression of UCP1 gene in adult abdominal fat has not been found to be similar to that of BMI, waist. Blood pressure and blood lipids were correlated to different degrees, but none of them reached significant level. Only a high negative correlation was found between UCP1 expression and insulin resistance index. It is well known that abdominal fat accumulation can induce insulin resistance. Our results suggest that the mechanism may be related to the presence and quantity of brown fat in abdominal adipose tissue. This conclusion needs further study and confirmation.
