人體腹部脂肪中解偶聯(lián)蛋白1(UCP1)基因表達(dá)水平的探索研究

【摘要】: 目的:寒冷環(huán)境中維持機(jī)體溫度或由食物能量引起營養(yǎng)過剩時,能量的消耗主要通過棕色脂肪組織的產(chǎn)熱功能。二者發(fā)生機(jī)制都源于一種特異的蛋白,偶聯(lián)蛋白1 (uncoupling protein 1,UCP1)。而UCP1僅在棕色脂肪細(xì)胞中的線粒體內(nèi)表達(dá)。本研究即是通過探索成人腹部脂肪(包括皮下和內(nèi)臟)中是否存在有UCP1,以此來驗(yàn)證成人體內(nèi)白色脂肪組織(white adipose tissue, WAT)中是否存在棕色脂肪組織(brown adipose tissue, BAT)。進(jìn)一步探討成人腹部皮下、內(nèi)臟脂肪組織中UCP1的基因表達(dá)水平與成人血脂指標(biāo)、體質(zhì)指數(shù)、腰圍、基礎(chǔ)代謝的相關(guān)關(guān)系。方法:研究樣本取自2008年8月~2009年1月期間在新疆維吾爾自治區(qū)人民醫(yī)院普外二科新入院且需進(jìn)行開腹外科手術(shù)的25例病人,經(jīng)簽署知情同意書后,在術(shù)中取得皮下和內(nèi)臟新鮮脂肪組織各3g。應(yīng)用RT-PCR技術(shù)檢測人體腹部脂肪組織中UCP1的基因是否存在表達(dá)及表達(dá)水平,以此研究棕色脂肪組織的基因表達(dá)與人體能量代謝和肥胖的相關(guān)關(guān)系。UCP1擴(kuò)增引物序列參考Genbank的已知序列(Gene ID:24860,|ref|NM_021833.3)設(shè)計(jì)。進(jìn)一步分析成人腹部脂肪組織中UCP1mRNA基因表達(dá)量與身高、體重、體質(zhì)指數(shù)、腰臀圍、收縮壓、舒張壓、空腹血漿血糖、總膽固醇、甘油三酯、高密度脂蛋白、胰島素抵抗指數(shù)及基礎(chǔ)代謝相關(guān)關(guān)系。結(jié)果:研究對象共25例(男性13例),平均年齡(44.68±10.93)歲。在25例研究對象中,21例成功實(shí)現(xiàn)擴(kuò)增,檢測皮下脂肪組織中UCP1/β-actin灰度值比值(0.79±0.12),內(nèi)臟脂肪組織中UCP1/β-actin灰度值比值(0.82±0.11),二者灰度值比值無統(tǒng)計(jì)學(xué)差異(P=0.822)。PCR擴(kuò)增產(chǎn)物測序結(jié)果與GenBank上已知序列比對有99%的同源性。人體腹部脂肪組織中UCP1基因表達(dá)量與體重指數(shù)(BMI)、身高、體重、腰圍、血壓、總膽固醇(TC)、甘油三酯(TG)、血糖(GLU)、高密度脂蛋白(HDL)、空腹血漿血糖(FPG)均存在不同程度的相關(guān)關(guān)系,但均未達(dá)到顯著性水平。僅發(fā)現(xiàn)胰島素抵抗指數(shù)(HOMA-IR)與UCP1基因表達(dá)量有高度的負(fù)相關(guān)關(guān)系(r=-0.700,P=0.036)。結(jié)論:本研究結(jié)果證實(shí)成人腹部脂肪中確實(shí)有棕色脂肪存在;棕色脂肪的表達(dá)量在成人腹部皮下與內(nèi)臟白色脂肪組織中沒有差別;此次研究沒有發(fā)現(xiàn)UCP1基因表達(dá)量與BMI、腰圍、血壓、血脂存在不同程度的相關(guān)關(guān)系,但均未達(dá)到顯著水平。僅發(fā)現(xiàn)UCP1表達(dá)量與胰島素抵抗指數(shù)呈現(xiàn)了高度的負(fù)相關(guān)。眾所周知,腹部脂肪堆積能夠引起的胰島素抵抗,本研究結(jié)果提示其機(jī)制可能與腹部脂肪組織中棕色脂肪的存在及其數(shù)量有關(guān)。該結(jié)論仍需進(jìn)一步研究證實(shí)。
[Abstract]:Aim: to maintain the body temperature in cold environment or to cause excess nutrition by food energy, the energy consumption is mainly through the heat production function of brown adipose tissue. Both mechanisms were derived from a specific protein, conjugate protein 1 (uncoupling protein 1 (UCP1). UCP1 was expressed only in mitochondria in brown adipocytes. This study was to investigate the presence of UCP1, in adult abdominal fat (including subcutaneous and visceral) to verify the presence of brown adipose tissue (brown adipose tissue, BAT). In white adipose tissue (white adipose tissue, WAT) in adults. To further investigate the relationship between UCP1 gene expression and lipid index, body mass index, waist circumference and basal metabolism in adult abdominal subcutaneous and visceral adipose tissue. Methods: from August 2008 to January 2009, 25 patients who were newly admitted to the second department of the people's Hospital of Xinjiang Uygur Autonomous region and who needed to undergo open surgery were studied. Subcutaneous and visceral fresh adipose tissue were obtained 3 g and 3 g respectively. RT-PCR technique was used to detect the expression and expression level of UCP1 gene in human abdominal adipose tissue. The relationship between gene expression in brown adipose tissue and energy metabolism and obesity was studied. UCP1 amplified primer sequence was designed to refer to the known Genbank sequence (Gene ID:24860, ref NM_021833.3). To further analyze the expression of UCP1mRNA gene in adult abdominal adipose tissue, including height, body weight, body mass index, waist-hip circumference, systolic blood pressure, diastolic blood pressure, fasting plasma glucose, total cholesterol, triglyceride, high density lipoprotein. Relationship between insulin resistance index and basal metabolism. Results: a total of 25 subjects (13 males) with an average age of (44.68 鹵10.93) years were enrolled in the study. Among the 25 subjects, 21 cases were successfully amplified. The gray value ratio of UCP1/ 尾 -actin in subcutaneous adipose tissue and visceral adipose tissue was (0.79 鹵0.12) and (0.82 鹵0.11), respectively. The expression of UCP1 gene in abdominal adipose tissue was correlated with body mass index (BMI),) height, weight, waist circumference, blood pressure, total cholesterol (TC), triglyceride, (TG), (GLU), high density lipoprotein (HDL), fasting plasma glucose and (FPG). But all did not reach the significant level. Only the insulin resistance index (HOMA-IR) was negatively correlated with the expression of UCP1 gene (r-0.700). Conclusion: the results of this study confirm the presence of brown fat in adult abdominal fat, the expression of brown fat in subcutaneous and visceral white adipose tissue of adult abdomen is not different, and the expression of UCP1 gene in adult abdominal fat has not been found to be similar to that of BMI, waist. Blood pressure and blood lipids were correlated to different degrees, but none of them reached significant level. Only a high negative correlation was found between UCP1 expression and insulin resistance index. It is well known that abdominal fat accumulation can induce insulin resistance. Our results suggest that the mechanism may be related to the presence and quantity of brown fat in abdominal adipose tissue. This conclusion needs further study and confirmation.
