大鼠慢性局灶性腦低灌注模型建立及神經(jīng)細(xì)胞凋亡的動(dòng)態(tài)觀測(cè)

【摘要】:目的: 本課題利用改良大鼠線栓法致MCAO建立慢性局灶性腦低灌注模型,研究慢性局灶性低灌注條件下神經(jīng)細(xì)胞形態(tài)學(xué)的改變及血清hs-CRP的變化,為研究腦血管疾病及神經(jīng)退行性疾病提供基礎(chǔ)實(shí)驗(yàn)依據(jù)。 方法: 1、成年健康SD雄性大鼠60只,隨機(jī)平均分為正常對(duì)照組、假手術(shù)組及造模后后1 d、3 d、7 d、14 d共6組,通過改良線栓法MCAO模型。應(yīng)用神經(jīng)行為學(xué)評(píng)分、TTC染色觀察腦片及光鏡、透射電鏡觀察細(xì)胞形態(tài)學(xué)的改變。 2、采用實(shí)驗(yàn)一所用大鼠。應(yīng)用免疫組化及ELISA方法,觀察rCCH大鼠不同時(shí)間點(diǎn)腦組織神經(jīng)細(xì)胞凋亡的表達(dá)及血清中hs-CRP的含量。 結(jié)果: 1、各組大鼠術(shù)后兩種神經(jīng)行為學(xué)評(píng)分均≤1分,各組相互比較差異無統(tǒng)計(jì)學(xué)意義(P0.05);對(duì)照組與實(shí)驗(yàn)組大鼠腦切片TTC染色均未見局灶性腦梗灶。電鏡可見神經(jīng)細(xì)胞胞漿內(nèi)細(xì)胞器崩解。 2、血清hs-CRP含量和神經(jīng)細(xì)胞凋亡檢測(cè):術(shù)后1 d、3 d、7 d時(shí),rCCH模型大鼠血清中hs-CRP含量與正常對(duì)照組比較,差異無顯著性(P0.05),術(shù)后14 d時(shí),rCCH模型大鼠血清中hs-CRP含量高于正常對(duì)照組,差異有顯著性(P0.05);但hs-CRP數(shù)值在隨日期遞增,由此說明,在術(shù)后1-2 w時(shí),隨著實(shí)驗(yàn)時(shí)間的延長(zhǎng),rCCH模型大鼠血清中hs-CRP含量會(huì)繼續(xù)增加。TUNEL法神經(jīng)細(xì)胞凋亡染色顯示,術(shù)后1 d內(nèi)神經(jīng)細(xì)胞凋亡細(xì)胞急劇增多,以星型膠質(zhì)細(xì)胞為著,1 d組、3 d組及對(duì)照組間相比(P0.01);7 d到14 d間凋亡細(xì)胞依然呈增多趨勢(shì),但是由于時(shí)間的間隔,增加的數(shù)目相對(duì)減少。 結(jié)論: 1、通過改良大鼠線栓致MCAO模型,可成功建立較穩(wěn)定的大鼠慢性局灶性低灌注模型。 2、慢性局灶性腦低灌注可導(dǎo)致供血區(qū)神經(jīng)細(xì)胞過度凋亡,但未見片狀炎性壞死。 3、rCCH大鼠MCA供血區(qū)神經(jīng)細(xì)胞凋亡隨術(shù)后時(shí)間的延長(zhǎng)而逐漸加重。
[Abstract]:Objective: to establish a chronic focal cerebral hypoperfusion model by using modified rat thread occlusion method to study the morphological changes of nerve cells and the changes of serum hs-CRP in chronic focal cerebral hypoperfusion. To provide basic experimental basis for the study of cerebrovascular diseases and neurodegenerative diseases. Methods: 1. Sixty adult healthy male SD rats were randomly divided into normal control group, sham operation group and 6 groups at 1 day, 3 days and 7 days and 14 days after modeling. The MCAO model was established by modified suture method. The neurobehavioral score and TTC staining were used to observe the brain slices and light microscope, and the morphological changes were observed by transmission electron microscope. The expression of neuronal apoptosis and the content of hs-CRP in serum of rCCH rats at different time points were observed by immunohistochemical and ELISA methods. Results: 1.
