大鼠骨移植癌痛模型的建立及其機(jī)制的初步研究

【摘要】: 癌癥痛嚴(yán)重影響病人生活質(zhì)量,其機(jī)制研究剛剛起步。本工作參照文獻(xiàn),通過(guò)測(cè)定模型大鼠體重,脛骨病理。脛骨X光片,后肢熱痛覺(jué)過(guò)敏,機(jī)械性痛覺(jué)超敏(觸誘發(fā)痛),自發(fā)性疼痛等綜合評(píng)估方法建立癌癥實(shí)驗(yàn)動(dòng)物模型。并運(yùn)用免疫組化等方法,觀察癌痛時(shí)大鼠脊髓背角膠質(zhì)細(xì)胞功能活動(dòng)變化,探討膠質(zhì)細(xì)胞在癌痛中的作用。結(jié)果提示脛骨內(nèi)接種walker256細(xì)胞可產(chǎn)生多因素依賴的疼痛行為學(xué)改變,并與臨床表現(xiàn)相吻合,適合作為癌癥性疼痛動(dòng)物模型用于癌癥性疼痛的機(jī)制及治療研究;大鼠脛骨癌痛時(shí),腫瘤細(xì)胞接種對(duì)側(cè)后肢也出現(xiàn)機(jī)械性痛覺(jué)超敏,提示該模型可能存在鏡像痛;癌痛時(shí)雙側(cè)脊髓背角星型膠質(zhì)細(xì)胞和小膠質(zhì)細(xì)胞的激活是雙側(cè)的,提示可能與鏡像痛有關(guān),并可能參與癌痛的過(guò)程,但是脊髓膠質(zhì)細(xì)胞在癌痛中的確切作用還有待于進(jìn)一步深入研究。
[Abstract]:Cancer pain seriously affects the quality of life of patients, its mechanism research has just started. In this work, the body weight and tibia pathology of the model rats were measured with reference to the literature. The animal model of cancer was established by comprehensive evaluation methods of tibia X-ray, thermal hyperalgesia in hind limbs, mechanical hyperalgesia (touch-induced pain) and spontaneous pain. The functional changes of glial cells in the spinal dorsal horn of rats were observed by immunohistochemical method, and the role of glial cells in cancer pain was discussed. The results suggest that walker256 cells inoculated in the tibia can produce multifactor dependent pain behavior changes, which are consistent with the clinical manifestations, and are suitable for the mechanism and treatment of cancer pain as an animal model of cancer pain. Mechanical pain hypersensitivity was also found in the contralateral hindlimb inoculated with tumor cells, suggesting that there might be mirror pain in the model, and that the activation of astrocytes and microglia in the dorsal horn of the spinal cord was bilateral, suggesting that it might be related to mirror pain. And may be involved in the process of cancer pain, but the exact role of spinal glial cells in cancer pain needs further study.
