低色氨酸培養(yǎng)基與色氨酸代謝產(chǎn)物聯(lián)合抑制T細胞增殖的體外研究

【摘要】: 目的:本課題應用小鼠骨髓來源樹突細胞(Dendritic Cell, DC)刺激同種異系小鼠T淋巴細胞,應用低色氨酸培養(yǎng)基及色氨酸代謝產(chǎn)物抑制T淋巴細胞增殖,并揭示其抑制T淋巴細胞增殖機制。 方法:采用培養(yǎng)基細胞因子選擇法體外培養(yǎng)小鼠骨髓來源DC,流式細胞儀檢測DC表面抗原CD80、CD86、MHCⅡ、CD11c表達情況,在加入色氨酸代謝產(chǎn)物的低色氨酸培養(yǎng)基中混合淋巴細胞培養(yǎng)并觀察對異系T淋巴細胞增殖能力的影響。利用Annexin-V及PI雙染法測定T細胞凋亡情況。使用SPSS 15.0軟件包進行數(shù)據(jù)分析。 結果:經(jīng)過體外培養(yǎng),每只小鼠可獲得5-6×106個骨髓來源DC,細胞具備典型樹突狀結構,高表達CD80 (94.56%)、CD86 (88.42%)、MHC-Ⅱ(86.12%)、CD11c(96.79%)。經(jīng)免疫磁珠法可分離出純度達90%以上的CD4+T細胞,進行混合淋巴細胞培養(yǎng),低色氨酸組較正常色氨酸組刺激指數(shù)(SI)明顯降低(P0.001),加入色氨酸代謝產(chǎn)物KYN組、3-HK組、3-HAA組SI值均隨其濃度升高而明顯降低,且與對照組相比具有統(tǒng)計學差異(P0.001)。其中以3-HAA抑制CD4+T細胞增殖作用最明顯,所需濃度最低。低色氨酸培養(yǎng)基與色氨酸代謝產(chǎn)物對CD4+T細胞增殖的抑制具有疊加作用。Annexin-V及PI雙染法測定各組CD4+T細胞凋亡低色氨酸組凋亡率明顯升高,且加入色氨酸代謝產(chǎn)物后凋亡率進一步升高(P0.001)。 結論:培養(yǎng)基細胞因子選擇法可收獲大量的骨髓源DC,免疫磁珠法可分離純度較高的CD4+T細胞。低色氨酸培養(yǎng)基及色氨酸代謝產(chǎn)物均可抑制CD4+T細胞增殖,并且二者具有疊加效應。其造成CD4+T細胞增殖抑制的機制為“色氨酸饑餓”和“色氨酸代謝產(chǎn)物毒性作用”造成細胞生長停滯甚至凋亡。這為進一步研究器官移植后的免疫耐受提供了實驗基礎。
