大鼠胎腦放射狀膠質(zhì)細(xì)胞表達(dá)乙酰膽堿轉(zhuǎn)移酶
[Abstract]:To confirm the expression of ChAT (acetylcholine transferase) in radiative glial cells of the dorsal ventricular region of the endencephalon to understand whether it produces cholinergic neurons. The morphology, cell division and expression of ChAT (acetylcholine transferase) of cortical pyramidal neurons produced at the dorsal end of the rat brain were observed by immunohistochemistry and electron microscopy. Firstly, Vimentin (vimentin) was used for immunohistochemical staining of cultured cells. The results showed positive expression. The primary culture of E14 rat embryonic brain cells was carried out in vitro. The cultured cells were stained with vimentin and acetylcholine transferase double fluorescent immunocytochemistry to determine whether the radial glial cells expressed acetylcholine transferase in vitro. The position of acetylcholinyltransferase (acetylcholinyltransferase) expressed by radial glial cells was precisely reflected by overlapping fluorescence monochromatic images in the same field. The results showed that Vimentin/ChAT immunocytochemical double staining was present in the cytoplasm of E14 rat embryonic brain cells. There were many double staining positive cells in the cell clusters, and the interlinked processes could be seen between the cell clusters. The cells migrated along the processes between the cell clusters, and there was Vimentin/ChAT immunocytochemical double staining positive expression in the processes. The results confirmed the expression of acetylcholine transferase in radiative glial cells of E14 rats in vitro. The results indicate that the radial glial cells have the nature of neural stem cells, and they divide unsymmetrically and produce cholinergic phenotypic neural cells, that is, cholinergic progenitor cells. The asymmetric division of radial glial cells during neuronogenesis has determined the phenotype of neurons and synthesized neurotransmitter acetylcholine. The results suggest that: (1) radiative glial cells can produce cholinergic neurons; (2) radial glial cells may be classified into different subgroups according to the types of neurons produced; (3) radial glial cells may have developed in the cerebral cortex.
