深圳市新分離兩株基孔肯雅熱病毒的分子遺傳學(xué)分析

【摘要】:目的對(duì)2010~2012年深圳市報(bào)告的兩起輸入性基孔肯雅熱病原體進(jìn)行分子遺傳學(xué)分析。方法利用C6/36細(xì)胞從病人血清中分離基孔肯雅病毒(Chikungunya virus,CHIKV),對(duì)分離得到的CHIKV進(jìn)行全基因組測(cè)序和構(gòu)建系統(tǒng)發(fā)生樹(shù),結(jié)合流行病學(xué)資料對(duì)其分子遺傳特征進(jìn)行分析。結(jié)果成功分離得到兩株CHIKV,分別命名為SZ_20101028和SZ_20120702,SZ_20101028全基因組長(zhǎng)為12377bp,SZ_20120823全基因組長(zhǎng)為11893bp;構(gòu)建系統(tǒng)發(fā)生樹(shù)分析,結(jié)果表明,SZ_20101028是印度洋亞型,ECSA型的后裔。SZ_20120702為Asian亞型。結(jié)論 SZ_20101028與最近10年來(lái)在印度洋島嶼爆發(fā)流行新亞型且與2010年由輸入性引發(fā)起東莞疫情爆發(fā)的毒株親源性最高,為99%;而SZ_20120702為Asian亞型,不是近年來(lái)爆發(fā)流行株。
[Abstract]:Objective to analyze the molecular genetics of two imported Kikungunya fever pathogens reported in Shenzhen from 2010 to 2012. Methods Chikungunya virus was isolated from patients' serum by C6 / 36 cells. The whole genome of CHIKV was sequenced and phylogenetic tree was constructed. The molecular genetic characteristics were analyzed with epidemiological data. Results two strains of CHIKVwere successfully isolated and named as SZ_20101028 and SZS _ (20120702) respectively. The length of the whole genome of SZ _ (201028) was 12377bpmpmp, and the whole genome length of SZ _ (20823) was 11893bp.The result of phylogenetic tree analysis showed that SZZ20101028 was a descendant of Indian Ocean subtype (SZ2020702) and was subtype of Asian (S / Z20120702), and the result of phylogenetic tree analysis showed that SZZ20101028 was a subtype of Indian Ocean subtype. Conclusion the new subtype of SZ_20101028 epidemic in the Indian Ocean islands in the last 10 years and that of Dongguan outbreak caused by import in 2010 are the highest (99%), while SZ_20120702 is a Asian subtype, and is not a prevalent strain in recent years.
