鋅轉(zhuǎn)運(yùn)體ZnT8在小鼠睪丸和卵巢中表達(dá)和分布的研究
[Abstract]:Zinc is one of the essential trace elements in the body, and it is necessary for many metal binding proteins and cells to function normally. Zinc ions can not cross the cell membrane freely, and their transmembrane transport requires specific zinc ion transporter to maintain the homeostasis of zinc ions in cells. Zinc transporter ZnT8 belongs to SLC30 family. It is cloned from SLC30A8 gene (chromosome 8q24.11) and is a protein containing 369 amino acid fragments. In the process of insulin synthesis, ZnT8, which exists on the insulin-secreting vesicles, makes zinc ions enter the vesicles to participate in the formation of zinc hexamers, and then participate in the process of insulin formation and storage in cells. Because ZnT8 is also expressed in islet alpha cells, thyroid acinar epithelial cells, adrenal cortical cells and other endocrine cells, ZnT8 is considered to be a zinc transporter closely related to endocrine cells. In this study, we investigated the expression of ZnT8 in endocrine cells of reproductive system and further confirmed the relationship between ZnT8 and endocrine cells. Materials and methods 30 healthy CD-1 adult mice (male and female, weighing 20-25 g) were selected and given sufficient food and tap water. Reverse transcriptase polymerase chain reaction (RT-PCR) was used to detect the expression of ZnT8 in testis and ovaries at mRNA level. The products of PCR were separated by 1% agarose gel electrophoresis. Immunohistochemical staining technique was used to detect the expression of ZnT8 in testis. The distribution of free zinc ions in testis and ovaries was demonstrated by (AMG) histochemical staining. The coexistence of ZnT8 and StAR protein in testicular Leydig cells and secondary follicular granulosa cells was confirmed by immunofluorescence staining. Results 1 RT-PCR results of mRNA expression in testis and ovaries RT-PCR showed that there were clear bands between 250bp and 500bp (the size of ZnT8 primer was 323bp). 2Immunohistochemical results of the distribution and expression of ZnT8 mRNA protein in testis were mainly located in testicular seminiferous cells compared with the negative control group. Leydig cells between tubes, At the same time, ZnT8 protein was also distributed in spermatogonia at the base of seminiferous tubules. 3. Zinc ions distributed in testis by AMG. The results showed that the distribution of zinc ions in testis tended to be concentrated towards the center of seminiferous tubules. In Leydig cells at the base of seminiferous tubules and between seminiferous tubules, The distribution of free zinc ions was also observed. 4. Zinc ions were distributed in the ovarian AMG results the free zinc ions of the ovary were widely distributed in the corpus luteum primary follicles and secondary follicles. In secondary follicles, A large number of zinc ions were distributed in oocytes and granulosa cells inside the follicular membrane. 5. ZnT8 immunofluorescence of testis Leydig cells and ovarian secondary follicles was found in Leydig cells of testis ZnT8 and StAR in granulosa cell layer of ovarian secondary follicles were detected by immunofluorescence double labeling. The results showed that ZnT8 and StAR proteins co-existed in these two steroid hormone secreting cells. Conclusion 1ZnT8 mRNA and protein were expressed in testis and ovary, 2the expression of ZnT8 was specific in testis. The expression of ZnT8 in testis and ovaries was mainly related to the location of StAR protein in Leydig cells.
