【摘要】：目的研究鳥結(jié)核分枝桿菌刺激巨噬細(xì)胞后其對巨噬細(xì)胞骨架蛋白及其調(diào)節(jié)蛋白的作用機制。方法 RT-PCR方法分析M.avium刺激巨噬細(xì)胞后cofilin-1,β-actin基因的表達水平,同時Western blot方法從蛋白水平分析M.avium刺激巨噬細(xì)胞后β-actin、cofilin-1蛋白的表達水平。流式細(xì)胞術(shù)檢測M.avium刺激巨噬細(xì)胞后巨噬細(xì)胞凋亡及壞死的情況。結(jié)果β-actin基因及其蛋白在巨噬細(xì)胞受M.avium刺激后表達顯著下調(diào),而β-actin蛋白的調(diào)節(jié)蛋白cofilin-1表達顯著增強。同時,M.avium刺激巨噬細(xì)胞后誘導(dǎo)巨噬細(xì)胞凋亡顯著增多。結(jié)論 M.avium刺激巨噬細(xì)胞后可通過調(diào)控β-actin及cofilin-1蛋白的表達,從而誘導(dǎo)巨噬細(xì)胞凋亡或壞死。
[Abstract]:Objective to study the effect mechanism of Mycobacterium tuberculosis on macrophage cytoskeleton and its regulatory protein after stimulating macrophages. Method RT-PCR method was used to analyze the expression level of cofilin-1, beta -actin gene after M.avium stimulation of macrophages, and Western blot method to analyze beta -actin, cofili after M.avium stimulation of macrophage from protein level. The expression level of n-1 protein. Flow cytometry was used to detect the apoptosis and necrosis of macrophage after M.avium stimulated macrophage. Results the expression of beta -actin gene and its protein decreased significantly after M.avium stimulation, while the expression of the regulatory protein of beta -actin protein was significantly enhanced. Meanwhile, M.avium stimulated macrophage to induce the expression. The apoptosis of macrophages was significantly increased. Conclusion M.avium stimulated macrophage by regulating the expression of beta -actin and cofilin-1 protein, thus inducing macrophage apoptosis or necrosis.
