【摘要】：目的獲得一種適于絲狀真菌基因研究用的、由根癌農(nóng)桿菌介導(dǎo)的RNA干擾方法。方法采用基因重組及菌株干擾體系篩選的方法。結(jié)果獲得適于根癌農(nóng)桿菌轉(zhuǎn)染的重組載體PCB309-pfgrt,并將其成功用于對(duì)孢子絲菌雙組份信號(hào)組氨酸蛋白激酶DRK1基因的干擾中。結(jié)論該方法優(yōu)化了根癌農(nóng)桿菌的轉(zhuǎn)化體系,解決了絲狀真菌RNA干擾載體構(gòu)建困難及干擾效率低下的難題,在絲狀真菌基因功能及遺傳轉(zhuǎn)化研究方面具有廣泛的應(yīng)用前景。
[Abstract]:Objective to obtain a method of RNA interference, which is suitable for the study of filamentous fungi gene, which is mediated by Agrobacterium tumefaciens. Methods the method of gene recombination and screening of strain interference system was used. The recombinant vector PCB309-pfgrt suitable for Agrobacterium tumefaciens transfection was obtained, and it was successfully applied to the double component signal histidine kinase D of sporomonas sp. In the interference of RK1 gene, conclusion this method optimizes the transformation system of Agrobacterium tumefaciens, and solves the difficult problem of the construction of filamentous fungi RNA interference carrier and the low interference efficiency. It has a wide application prospect in the study of the gene function and genetic transformation of filamentous fungi.
