【摘要】：目的:建立適宜于抗抑郁藥物評價的抑郁大鼠動物模型及綜合評價體系,為抗抑郁藥物的藥理藥效評價奠定前期基礎(chǔ)。方法:選用SD雄性大鼠,在CUMS法基礎(chǔ)上,建立抑郁大鼠動物模型,考察造模前和造模7天、14天、21天、35天時大鼠體重、懸尾次數(shù)、糖水消耗、血清皮質(zhì)酮等指標(biāo)的變化,綜合評價優(yōu)選造模條件。結(jié)果:與空白組比較,造模21天模型組在體重、糖水消耗、懸尾次數(shù)、游泳時間、敞箱實驗等方面均有明顯降低趨勢,大鼠血清皮質(zhì)酮含量明顯減少,實驗結(jié)果證明了抗抑郁藥物評價的抑郁大鼠動物模型建立的合理性。結(jié)論:該模型克服以往存在的工作量大,持續(xù)時間長、指標(biāo)評價單一等問題,為抗抑郁藥物的藥效評價奠定實驗基礎(chǔ)。
[Abstract]:Aim: to establish an animal model and a comprehensive evaluation system for antidepressant drug evaluation in rats, so as to lay a foundation for the evaluation of pharmacological efficacy of antidepressant drugs. Methods: SD male rats were used to establish the depressive rat model on the basis of CUMS method. The changes of body weight, tail suspension times, sugar consumption and serum corticosterone were investigated before and after 7 days and 14 days and 21 days and 35 days after the establishment of the model. Comprehensive evaluation of optimal model making conditions. Results: compared with the blank group, the body weight, sugar water consumption, tail suspension times, swimming time, open box test and serum corticosterone content in the 21-day model group were significantly decreased. The results demonstrated the rationality of the animal model of antidepressant drug evaluation in depressive rats. Conclusion: this model overcomes the problems of heavy workload, long duration and single index evaluation, which can be used to evaluate the efficacy of antidepressants.
