江西地區(qū)漢族人群6個(gè)STR基因座位群體遺傳多態(tài)性研究

【摘要】: 目的:調(diào)查江西地區(qū)漢族人群無關(guān)個(gè)體D6S1043、D2S1772、D7S3048、D22-GATA198B05、D8S1132與D11S2368等6個(gè)STR基因座的遺傳多態(tài)性,積累群體遺傳學(xué)數(shù)據(jù),探討其應(yīng)用價(jià)值。 方法:采集江西地區(qū)212例漢族無關(guān)個(gè)體EDTA抗凝全血,應(yīng)用STR-PCR與PAGE檢測D6S1043、D2S1772、D7S3048、D22-GATA198B05、D8S1132與D11S2368等6個(gè)STR基因座的等位基因。 結(jié)果:在212例江西地區(qū)漢族人群中觀察到D6S1043基因座位13個(gè)基因,52種基因型;D2S1772基因座13種基因,66種基因型;D7S3048基因座位12種基因,48種基因型;D22-GATA198B05基因座位11種基因,44種基因型;D8S1132基因座10種基因,38種基因型;D11S2368基因座位10種基因41種基因型。D6S1043、D2S1772、D7S3048、D22-GATA198B05、D11S2368與D8S1132等6個(gè)基因座的觀察雜合度(h)分別為:0.8019、0.8302、0.8538、0.8302、0.8774與0.8443.期望雜合度(H)分別為:0.8727、0.8896、0.8813、0.8553、0.8559、0.8611;個(gè)人識(shí)別率(DP)分別為:0.9662、0.9735、0.9697、0.9607、0.9559、0.9613;非父排除率(PE)分別為:0.7394、0.7751、0.7445、0.7086、0.7053、0.7219;多態(tài)信息含量(PIC)分別為:0.8475、0.8774、0.8730、0.8453、0.8452、0.8510。6個(gè)位點(diǎn)的累積非父排除率(CPE)為0.0000000001,累積個(gè)人識(shí)別率(CDP)為0.9996423770。 結(jié)論:本次調(diào)查6個(gè)STR位點(diǎn)在江西地區(qū)漢族人群的分布符合Hardy-Weinberg平衡,遺傳多態(tài)性高。本次調(diào)查為豐富江西地區(qū)遺傳資料庫及實(shí)現(xiàn)本地區(qū)法醫(yī)學(xué)親子鑒定和個(gè)人識(shí)別提供了基礎(chǔ)數(shù)據(jù)和理論依據(jù)。
[Abstract]:Objective: To investigate the genetic polymorphism of 6 independent STR loci, such as D6S1043, D2S1772, D7S3048, D22-GATA198B05, D8S1132 and D11S2368, in the Han population of Jiangxi, and to accumulate the population genetic data and explore its application value.
Methods: EDTA anticoagulant blood was collected from 212 unrelated individuals of Han nationality in Jiangxi. The alleles of 6 STR genes, such as D6S1043, D2S1772, D7S3048, D22-GATA198B05, D8S1132 and D11S2368, were detected by STR-PCR and PAGE.
Results: 13 genes, 52 genotypes, 13 genes, 66 genotypes, 12 D7S3048 gene seats, 48 genotypes, 11 gene loci, 44 genotypes of D22-GATA198B05, 10, 38 and D11S2368 genes of the D8S1132 loci were observed in 212 Jiangxi Han population. The heterozygosity (H) of 6 loci.D6S1043, D2S1772, D7S3048, D22-GATA198B05, D11S2368 and D8S1132, respectively, were: 0.8019,0.8302,0.8538,0.8302,0.8774 and 0.8443. expected heterozygosity (H) respectively: 0.8727,0.8896,0.8813,0.8553,0.8559,0.8611, and the individual recognition rate (DP) was respectively: 0.9607,0.9559,0.9613, the non parent exclusion rate (PE) was 0.7394,0.7751,0.7445,0.7086,0.7053,0.7219, and the polymorphic information content (PIC) was: the cumulative non parent exclusion rate (CPE) of 0.8475,0.8774,0.8730,0.8453,0.8452,0.8510.6 sites was 0.0000000001, and the cumulative individual recognition rate (CDP) was 0.9996423770..
Conclusion: the distribution of 6 STR loci in the Jiangxi Han population in this survey accords with the balance of Hardy-Weinberg and high genetic polymorphism. This survey provides basic data and theoretical basis for enriching the genetic database in Jiangxi area and realizing the paternity and personal identification of forensic medicine in the region.
