【摘要】:[目的]用編碼單純皰疹病毒Ⅰ型(HSV-1)糖蛋白B的pDNA轉(zhuǎn)染小鼠,研究基因疫苗治療感染性疾病的有效性,并分析免疫反應(yīng).[方法]采用血管內(nèi)注射方法,將含有不同劑量、不同pDNAs的生理鹽水注入Balb/c小鼠體內(nèi),1周后行再次免疫,觀察以不同組合pDNA進(jìn)行免疫的實(shí)驗(yàn)組和對(duì)照組小鼠被致死劑量HSV-1感染后生存率間的差異.通過潛伏感染的檢測(cè)、血清中細(xì)胞因子水平的測(cè)定、細(xì)胞毒性反應(yīng)的測(cè)定及中和實(shí)驗(yàn)評(píng)價(jià)免疫效果.[結(jié)果]接種pG.gB或接種pGEG.gB的小鼠,與相應(yīng)對(duì)照組比較,生存率和誘導(dǎo)產(chǎn)生中和抗體的效價(jià)與劑量呈正相關(guān),pGEG.gB免疫組對(duì)P815.gB細(xì)胞有顯著的細(xì)胞毒性作用,pGEG.gB免疫小鼠的脾細(xì)胞增殖反應(yīng)顯著強(qiáng)于pG.gB免疫的小鼠;pG.gB和pGEG.gB免疫后的小鼠血清中IL-12和IFN-γ水平均顯著升高.[結(jié)論]裸pDNA的經(jīng)小鼠尾靜脈免疫可誘導(dǎo)HSV-1感染小鼠的體液和細(xì)胞免疫應(yīng)答,對(duì)小鼠HSV-1急性感染有明顯的預(yù)防和治療作用,且含有EBNA1和oriP的EBV質(zhì)粒載體明顯增強(qiáng)CTL活性和增殖反應(yīng).
[Abstract]:[objective] to study the efficacy of gene vaccine in the treatment of infectious diseases by transfection of pDNA encoding herpes simplex virus type I (HSV-1) glycoprotein B in mice, and to analyze the immune response. [methods] by intravascular injection, saline containing different doses and different pDNAs was injected into Balb / c mice for 1 week and then immunized again. The survival rate of mice immunized with different combinations of pDNA by lethal dose HSV-1 was observed. The immune effect was evaluated by the detection of latent infection, the level of cytokines in serum, the determination of cytotoxic reaction and neutralization experiment. [results] the mice inoculated with pG.gB or pGEG.gB were compared with the control group. Survival rate and the titer of inducing neutralizing antibody were positively correlated with the dose of pGEG.gB immunized mice. The cytotoxicity of pGEG.gB immunized mice to P815.gB cells was significantly stronger than that of pGEG.gB immunized mice after pG.gB and pGEG.gB immunized mice. The levels of IL-12 and IFN- 緯 in serum of mice were significantly increased. [conclusion] Naked pDNA immunization via tail vein can induce humoral and cellular immune responses in mice infected with HSV-1, and has obvious preventive and therapeutic effects on acute HSV-1 infection in mice. And EBV plasmid vector containing EBNA1 and OLP significantly enhanced CTL activity and proliferation response.
