腹腔注射脂多糖聯(lián)合缺氧在腦癱模型評(píng)價(jià)的實(shí)驗(yàn)研究

【摘要】: 目的:探討孕鼠宮內(nèi)感染合并缺氧致幼鼠腦癱的模型制作及評(píng)價(jià)方法。方法:實(shí)驗(yàn)動(dòng)物分為三組:(1)脂多糖+缺氧組:選用孕16~18天健康Wistar雌性大鼠脂多糖腹腔注射(0.4mg/kg),12小時(shí)后置于缺氧環(huán)境中缺氧2.5小時(shí),4小時(shí)后再次腹腔注射脂多糖(0.4mg/kg);(2)頸動(dòng)脈結(jié)扎+缺氧組:組出生后第7天Wistar幼鼠結(jié)扎右側(cè)頸動(dòng)脈結(jié)合缺氧環(huán)境的方法制作幼鼠腦癱模型;(3)對(duì)照組:僅行頸正中切口,然后縫合切口。于術(shù)后4周對(duì)三組實(shí)驗(yàn)動(dòng)物進(jìn)行足印分析試驗(yàn)、股四頭肌復(fù)合肌肉運(yùn)動(dòng)誘發(fā)電位及病理學(xué)檢測(cè)。結(jié)果:(1)足印分析試驗(yàn):脂多糖+缺氧組和頸動(dòng)脈結(jié)扎+缺氧組實(shí)驗(yàn)鼠前后肢足印重復(fù)性差,表現(xiàn)為間距較大,且間距不穩(wěn)定,明顯大于對(duì)照組(P0.05);(2)兩實(shí)驗(yàn)組動(dòng)物后肢股四頭肌CMAP與對(duì)照組相比,引出最大波幅所需的刺激強(qiáng)度增大,引出波幅降低(P0.05); (3)HE染色光鏡觀察兩實(shí)驗(yàn)組腦室旁正常組織結(jié)構(gòu)破壞,細(xì)胞排列紊亂,局部大小不等圓形軟化灶,在頸動(dòng)脈結(jié)扎+缺氧組以損傷側(cè)腦室周圍變化明顯。結(jié)論:應(yīng)用孕鼠宮內(nèi)感染合并缺氧的方法制作幼鼠腦癱模型穩(wěn)定可靠,此模型與成熟的單側(cè)頸動(dòng)脈結(jié)扎缺氧模型相比具有微創(chuàng)的、簡(jiǎn)單易行、逼真的模擬人類在懷孕晚期宮內(nèi)感染缺氧致胎兒腦部損傷的優(yōu)點(diǎn),值得推廣。
[Abstract]:Objective: to investigate the model of cerebral palsy induced by intrauterine infection and hypoxia in pregnant rats. Methods: the experimental animals were divided into three groups: lipopolysaccharide hypoxia group: lipopolysaccharide (LPS) was injected intraperitoneally into female Wistar rats at gestation 1618 days after 12 hours of hypoxia for 2.5 hours and 4 hours. Artery ligation hypoxia group: right carotid artery ligation combined with anoxic environment in Wistar young rats on the 7th day after birth (n = 3) the model of cerebral palsy in young rats was established by anoxia) control group: the median cervical incision was used only, the right carotid artery was ligated on the 7th day after birth. Then suture the incision. Four weeks after operation, three groups of experimental animals were tested with foot print analysis, quadriceps femoris combined muscle motor evoked potential and pathological examination. Results (1) footprint analysis test: rats in lipopolysaccharide hypoxia group and carotid artery ligation hypoxia group had poor reproducibility of foreand hindlimb footprints, which were characterized by large spacing and unstable spacing. Compared with the control group, CMAP of the quadriceps femoris of the hind limbs of the two experimental groups increased the stimulation intensity needed to elicit the maximum wave amplitude, and decreased the amplitude of the CMAP, and the damage of the normal tissue structure around the ventricle of the two experimental groups was observed under light microscope. The cells were disordered, the local size was not equal to round softening foci, and the changes were obvious in the group of carotid artery ligation and hypoxia. Conclusion: the model of cerebral palsy in young rats made by intrauterine infection combined with hypoxia in pregnant rats is stable and reliable. Compared with the mature unilateral carotid artery ligation hypoxia model, this model is minimally invasive and easy to perform. The advantages of anoxic fetal brain injury caused by anoxia in late pregnancy are worth popularizing.
