【摘要】： 目的:研究低能量半導(dǎo)體激光對(duì)體外培養(yǎng)嗅粘膜嗅鞘細(xì)胞增殖的影響。方法:取成年SD大鼠嗅粘膜,采用差速貼壁法獲得嗅鞘細(xì)胞,對(duì)細(xì)胞進(jìn)行不同能量密度的激光照射(在10.3mW /cm2功率密度下分別照射30秒、60秒、120秒,每日一次,連續(xù)三天),采用細(xì)胞計(jì)數(shù)法和MTT法分別檢測(cè)照射后不同時(shí)間細(xì)胞增殖的變化。結(jié)果:培養(yǎng)3天時(shí),不同時(shí)間照射組和對(duì)照組細(xì)胞增殖無(wú)差異。培養(yǎng)5天和8天時(shí),不同時(shí)間照射組細(xì)胞數(shù)均高于對(duì)照組(p0.05),有統(tǒng)計(jì)學(xué)意義;60秒和120秒照射組OD值高于對(duì)照組(p0.05),有統(tǒng)計(jì)學(xué)意義;但30秒照射組OD值與對(duì)照組無(wú)差異。其中以60秒照射組細(xì)胞增值最為明顯。結(jié)論:低能量激光對(duì)嗅粘膜嗅鞘細(xì)胞的增殖有促進(jìn)作用并且與照射的能量密度有關(guān)。最佳照射時(shí)間是每日一次,連續(xù)三天。
[Abstract]:Aim: to study the effect of low energy semiconductor laser on the proliferation of olfactory ensheathing cells in vitro. Methods: olfactory ensheathing cells were obtained from olfactory mucosa of adult Sprague-Dawley rats. The cells were irradiated with laser with different energy density (30 seconds, 60 seconds, 120 seconds, once a day, respectively) at 10.3mW / cm2 power density. Cell count method and MTT method were used to detect cell proliferation at different time after irradiation for three consecutive days. Results: there was no difference in cell proliferation between irradiation group and control group at 3 days. At 5 and 8 days, the number of cells in the irradiation group was higher than that in the control group (P 0.05), and the OD value in the irradiation group at 60 seconds and 120 seconds was higher than that in the control group (P 0.05), but there was no difference in OD value between the 30 second group and the control group. In the 60 second group, the cell proliferation was the most obvious. Conclusion: low energy laser can promote the proliferation of olfactory ensheathing cells of olfactory mucosa and is related to the energy density of irradiation. The best exposure time is once a day for three consecutive days.
