局部亞低溫對大鼠不同腦缺血模型自體神經(jīng)干細(xì)胞的影響

【摘要】:目的:本課題通過觀察研究局部亞低溫治療對不同腦缺血模型自體神經(jīng)干細(xì)胞的影響,為今后干細(xì)胞的聯(lián)合治療提供實(shí)驗(yàn)依據(jù)。 方法:108只SD大鼠分為假手術(shù)、缺血再灌注(tMACO)、永久阻塞(pMACO)、tMACO+亞低溫、pMACO+亞低溫五組,各組根據(jù)時(shí)間點(diǎn)分為缺血6h、2d、7d三個(gè)亞組。線栓法建立大鼠腦缺血模型,亞低溫組栓塞成功后即刻予3h的頭部亞低溫治療(32-35℃)。記錄治療過程中動物直腸溫度,評估缺血前、缺血后6h、2d、7d動物一般情況、神經(jīng)功能缺損評分、梗死體積及brdu+、nestin+、brdu+/nestin+細(xì)胞數(shù)。 結(jié)果:術(shù)后6h,tMACO組神經(jīng)功能缺損評分明顯高于tMACO+亞低溫組,p=0.039,pMACO+亞低溫組明顯高于tMACO+亞低溫組,p=0.027;各實(shí)驗(yàn)組術(shù)后7d的神經(jīng)功能缺損評分均低于術(shù)后6h,p0.05;各實(shí)驗(yàn)組不同時(shí)間點(diǎn)梗死體積無差異;各實(shí)驗(yàn)組brdu+、nestin+、 brdu+/nestin+細(xì)胞于術(shù)后2天開始增加,術(shù)后7天的陽性細(xì)胞數(shù)目明顯高于術(shù)后6h,p0.05,而實(shí)驗(yàn)組之間陽性細(xì)胞數(shù)目差異無統(tǒng)計(jì)學(xué)意義;tMACO+亞低溫組術(shù)后2d、7d皮層中nestin+細(xì)胞數(shù)高于tMACO組,pMACO+亞低溫組術(shù)后2d皮層中nestin+細(xì)胞數(shù)高于tMACO組。 結(jié)論:腦缺血損傷可刺激自體神經(jīng)干細(xì)胞增殖;短暫輕度亞低溫治療可促進(jìn)自體神經(jīng)干細(xì)胞向損傷區(qū)遷徙;亞低溫聯(lián)合再灌注的腦保護(hù)作用優(yōu)于亞低溫聯(lián)合永久缺血,但優(yōu)勢短暫。圖12幅,表7個(gè),參考文獻(xiàn)53篇。
[Abstract]:Objective: to study the effect of local mild hypothermia on neural stem cells in different cerebral ischemia models, and provide experimental evidence for the combined treatment of stem cells in the future.
