彈性蛋白酶誘導(dǎo)兔囊性動(dòng)脈瘤模型的建立及其病理學(xué)研究

【摘要】: 目的:應(yīng)用豬胰彈性蛋白酶消化法建立兔囊性動(dòng)脈瘤模型,觀察該模型的形態(tài)學(xué)和病理學(xué)變化。 方法:應(yīng)用豬胰彈性蛋白酶消化頸總動(dòng)脈建立兔右頸總動(dòng)脈起始部的囊性動(dòng)脈瘤模型, MRA檢查動(dòng)態(tài)觀察該模型3天、14天、28天動(dòng)脈瘤形態(tài)學(xué)變化, HE染色和彈性纖維EVG染色法,光鏡和電鏡下觀察該模型0天、14天、28天動(dòng)脈瘤的病理學(xué)變化。 結(jié)果:動(dòng)脈瘤模型制作成功后早期動(dòng)脈瘤進(jìn)行性增大,14天后大小趨于穩(wěn)定。動(dòng)脈瘤模型病理學(xué)觀察發(fā)現(xiàn)0天動(dòng)脈瘤模型血管內(nèi)皮細(xì)胞消失,瘤壁彈力層完全缺失, 28天后動(dòng)脈瘤壁逐漸增厚,仍缺乏彈力層,但膠原纖維增生。 結(jié)論:豬胰彈性酶消化法建立的兔囊性動(dòng)脈瘤模型,不僅形態(tài)和病理上與人類顱內(nèi)動(dòng)脈瘤相似,而且早期具有自發(fā)生長的特性,提示該模型不僅可用于研究血管內(nèi)栓塞材料的實(shí)驗(yàn)研究,也可用于動(dòng)脈瘤形成機(jī)制的研究。
[Abstract]:Objective : To establish a model of rabbit cystic aneurysm by porcine pancreatic elastase digestion and to observe the morphological and pathological changes of the model .
Methods : The saccular aneurysm model of right common carotid artery was established with porcine pancreatic elastase . MRA was used to observe the morphological changes of aneurysm morphology , HE staining and EVG staining in the model , HE staining and EVG staining . The pathological changes of aneurysm were observed on 0 day , 14 days and 28 days under light microscope and electron microscope .
Results : The aneurysm model was more progressive after successful operation , and the size of aneurysm was stable after 14 days . The aneurysm model pathology observed that the vascular endothelial cells disappeared and the elastic layer of tumor wall was completely absent . After 28 days , the wall of aneurysm was gradually thickened , and the elastic layer was still lacking , but the collagen fibers were proliferated .
Conclusion : The rabbit cystic aneurysm model established by porcine pancreatic elastase is not only similar to human intracranial aneurysm but also spontaneous growth in early stage , suggesting that the model not only can be used to study the experimental study of endovascular embolization material , but also can be used in the study of aneurysm formation mechanism .
