【摘要】:目的:探討小鼠神經(jīng)干細(xì)胞(NSCs)免疫原性,以利于解決神經(jīng)干細(xì)胞移植中的免疫排斥問題。方法:(1)首先將30只C57BL/6近交系小鼠隨機分成兩組,以BALB/c近交系小鼠的NSCs及肝細(xì)胞(作為體細(xì)胞對照組)分別進(jìn)行腹腔主動免疫,然后將NSCs及肝細(xì)胞分別與各自免疫后小鼠的T淋巴細(xì)胞進(jìn)行單向混合淋巴細(xì)胞培養(yǎng),通過液閃計數(shù)儀檢測各自T淋巴細(xì)胞增殖程度,從而比較兩者免疫原性的強弱。(2)利用異硫氰酸熒光素標(biāo)記抗體(FITC)及Phycoery-thrin(PE)抗體分別標(biāo)記BALB/c小鼠NSCs主要組織相容性抗原復(fù)合物Ⅰ、Ⅱ(MHC-Ⅰ、MHC-Ⅱ),然后通過流式細(xì)胞術(shù)(FCM)分別檢測BALB/c小鼠NSCs及其肝細(xì)胞的MHC-Ⅰ和MHC-Ⅱ陽性細(xì)胞比例,從而推斷小鼠NSCs的免疫原性。結(jié)果:(1)NSC組液閃計數(shù)儀檢測所得cpm值為16592.8±2865.3,肝細(xì)胞組為27815.0±2416.3,NSC組明顯低于肝細(xì)胞組,差別有統(tǒng)計學(xué)意義(P0.01)。(2)小鼠NSCs實驗組中,約(87.55±3.65)%的增殖期神經(jīng)干細(xì)胞沒有檢測到MHC-Ⅰ和MHC-Ⅱ分子表達(dá);而作為對照組的小鼠肝細(xì)胞組中,僅有約(27.45±1.86)%的肝細(xì)胞未表達(dá)MHC-Ⅰ和MHC-Ⅱ分子。且NSC組MHC-Ⅰ和MHC-Ⅱ陰性細(xì)胞比例明顯高于肝細(xì)胞組,差別有統(tǒng)計學(xué)意義(P0.01)。結(jié)論:小鼠神經(jīng)干細(xì)胞具有較弱的免疫原性,且其免疫原性明顯低于同一個體的肝細(xì)胞。
[Abstract]:Aim: to investigate the immunogenicity of neural stem cells (NSCs) in mice in order to solve the problem of immune rejection in neural stem cell transplantation. Methods at first, 30 C57BL/6 inbred mice were randomly divided into two groups. The NSCs of BALB/c inbred mice and hepatocytes (as control group) were immunized by intraperitoneal active immunization. Then NSCs and hepatocytes were cultured with the T lymphocytes of mice immunized with each other, and the proliferation degree of T lymphocytes was measured by liquid scintillation counter.
