BAMBI基因過表達人結(jié)腸癌細胞移植斑馬魚致肝轉(zhuǎn)移模型的構(gòu)建

【摘要】:目的構(gòu)建BAMBI基因過表達人結(jié)腸癌細胞移植斑馬魚肝轉(zhuǎn)移模型,分析斑馬魚肝內(nèi)轉(zhuǎn)移性結(jié)腸癌細胞BAMBI基因表達水平與肝轉(zhuǎn)移發(fā)生率的關(guān)聯(lián)性。方法分別構(gòu)建BAMBI過表達穩(wěn)轉(zhuǎn)結(jié)腸癌細胞株SW620(BAMBI+組)、空載體紅色熒光蛋白基因轉(zhuǎn)染結(jié)腸癌細胞株SW620(EV組),分別顯微注射到受精后第2天轉(zhuǎn)基因斑馬魚Tg(Apo14∶GFP)胚胎卵黃囊(每組各680條),激光共聚焦熒光顯微鏡觀察、拍攝,采用Image J軟件測量轉(zhuǎn)移至斑馬魚肝臟內(nèi)結(jié)腸癌細胞紅色熒光表達強度,計算肝轉(zhuǎn)移發(fā)生率;采用t檢驗比較BAMBI+組及EV組斑馬魚肝臟內(nèi)結(jié)腸癌細胞紅色熒光表達強度及肝轉(zhuǎn)移發(fā)生率的差異。結(jié)果與EV組相比,激光共聚焦熒光顯微觀察顯示BAMBI+組斑馬魚肝內(nèi)結(jié)腸癌細胞紅色熒光表達量顯著增加(t=6.247,P=0.003 3);BAMBI+組斑馬魚肝轉(zhuǎn)移發(fā)生率與EV組相比顯著增加(t=4.276,P=0.012 9)。結(jié)論構(gòu)建了基因修飾人結(jié)腸癌細胞移植斑馬魚致肝轉(zhuǎn)移模型,可較好地模擬活體狀態(tài)的結(jié)腸癌肝轉(zhuǎn)移;BAMBI基因可促進異種移植入斑馬魚的結(jié)腸癌細胞發(fā)生肝轉(zhuǎn)移。
[Abstract]:Objective to establish a model of liver metastasis in zebrafish transplanted with human colon cancer cells with BAMBI gene overexpression, and to analyze the relationship between the expression of BAMBI gene and the incidence of liver metastasis in metastatic colon cancer cells of zebrafish. Methods BAMBI overexpression in colon cancer cell line SW620(BAMBI group was constructed, and empty vector red fluorescent protein gene was transfected into colon cancer cell line SW620(EV group. The embryo yolk sac of transgenic zebrafish TgApo14: GFP was microinjected into the embryo on the second day after fertilization. (680 pieces of each group were observed by laser confocal fluorescence microscope) Image J software was used to measure the expression intensity of red fluorescence in colon cancer cells transferred to zebrafish liver, and the incidence of liver metastasis was calculated. T test was used to compare the expression intensity of red fluorescence and the incidence of liver metastasis in BAMBI group and EV group. Results compared with EV group, laser confocal fluorescence microscopy showed that the expression of red fluorescence in colon cancer cells of zebrafish in BAMBI group was significantly higher than that in EV group. The incidence of hepatic metastasis of zebrafish in BAMBI group was significantly higher than that in EV group. Conclusion the model of liver metastasis induced by gene modified human colon cancer cell transplantation in zebrafish is constructed, which can simulate the liver metastasis of colon cancer cells in vivo and can promote liver metastasis of colonic cancer cells transplanted into zebrafish xenografts.
