兩種脾虛證模型對小鼠骨骼肌形態(tài)和功能的影響

【摘要】:目的探討脾虛證與骨骼肌收縮功能的關(guān)系。方法將36只ICR小鼠按體重配對原則隨機分成3組,即對照組、勞倦過度組及大黃組,每組12只。其中勞倦過度組及大黃組應用大黃瀉下法和力竭游泳加剝奪睡眠的方法制備兩種脾虛證動物模型;采用m-ATPase酶組織化學方法測定3組小鼠趾長伸肌(extensor digitorum longus,EDL)和比目魚肌(soleus,SOL)m-ATPase酶活性的變化,計算Ⅰ型和Ⅱ型肌纖維的比例;Power Lab系統(tǒng)測定EDL和SOL等長收縮和強直收縮的最大張力。結(jié)果勞倦過度組及大黃組小鼠的體重、體溫與一般健康狀況均明顯下降;脾臟、胸腺臟器指數(shù)亦低于對照組;等長收縮最大張力和強直收縮最大張力明顯下降;EDL和SOL的肌纖維橫截面積減小,細胞排列松散;EDL中Ⅰ型肌纖維比例增加,Ⅱ型肌纖維比例降低,SOL中Ⅰ型和Ⅱ型肌纖維比例無變化。結(jié)論勞倦過度組及大黃組小鼠的EDL和SOL均出現(xiàn)明顯萎縮,勞倦過度組SOL中Ⅰ型肌纖維萎縮更加明顯。
[Abstract]:Objective to study the relationship between spleen deficiency syndrome and skeletal muscle contraction function. Methods Thirty-six ICR mice were randomly divided into 3 groups according to the principle of body weight pairing: control group, overworked group and rhubarb group, 12 mice in each group. Two kinds of spleen deficiency syndrome animal models were established in the overworked group and rhubarb group by the method of rhubarb diarrhea and exhaustive swimming plus sleep deprivation. The enzyme activities of extensor digitorum longus (EDL) and soleus muscle (Soleusus soleus) were determined by m-ATPase histochemical method.
