廣西10例人芽囊原蟲基因型和同工酶譜的研究

【摘要】:目的分析廣西10個人芽囊原蟲(Blastocystis hominis)分離株基因型,及其乳酸脫氫酶(LDH)和酯酶(EST)的同工酶譜特征。方法從感染者糞便中分離到的10個人芽囊原蟲(BhGX1~BhGX10),體外培養(yǎng)并提取基因組DNA。用已知的7對特異性序列標(biāo)記位點(diǎn)(sequene tagged sites,STS)引物PCR擴(kuò)增,來鑒定基因型。采用十二烷基硫酸鈉-聚丙烯酰胺凝膠電泳(SDS-PAGE),分別進(jìn)行乳酸脫氫酶和酯酶2種同工酶染色,比較分析10個分離株的酶譜。結(jié)果 10個人芽囊原蟲分離株中有8個為基因Ⅰ型;另外2個分離株(BhGX4和BhGX7)經(jīng)7對引物擴(kuò)增均為陰性,為未知基因型。10個人芽囊原蟲分離株在LDH譜中共出現(xiàn)10條酶帶,常見酶帶為Rm37、Rm49、Rm57、Rm68和Rm92;在EST譜中共出現(xiàn)12條酶帶,其中Rm14、Rm18、Rm23、Rm27、Rm45、Rm50和Rm77酶帶較常見。各分離株之間的LDH和EST同工酶譜均存在差異。結(jié)論廣西10個人芽囊原蟲分離株以基因Ⅰ型為主,但各分離株之間的LDH和EST同工酶譜存在差異。
[Abstract]:Objective to analyze the isozyme patterns of lactic dehydrogenase (LDH) and esterase (EST) of 10 isolates of Blastocystis hominis from Guangxi. Methods BhGX1 (BhGX1) BhGX10 was isolated from the feces of infected individuals, and the genomic DNA was cultured and extracted in vitro. The genotypes were identified by PCR amplification using 7 pairs of known primers for sequene tagged sitesmotif. Two isozymes of lactate dehydrogenase and esterase were stained by sodium dodecyl sulfate and polyacrylamide gel electrophoresis (SDS-PAGEG). The zymograms of 10 isolates were compared and analyzed.
